Genetic, epigenetic, and environmental mechanisms govern allele-specific gene expression

Abstract: Allele-specific expression (ASE) is a phenomenon where one allele is preferentially expressed over the other. Genetic and epigenetic factors cause ASE by altering the final allelic composition of a gene's product, leading to expression imbalances that can have functional consequences on phenotypes. Environmental signals also impact allele-specific gene regulation, but how they contribute to this crosstalk remains understudied. Here, we explored how allelic genotype, parent-of-origin, tissue type, sex, and diet… Show more

“…Also for the African MHC class II, we observed large expression differences among putative loci with variation both within and among species. “…For example, plasticity in expression owing to cis -acting changes tend to accumulate in Arabidopsis to magnify pre-existing plasticity of expression [ 83 ]. Emerging studies that employ allele-specific expression to detect biased expression among a wide array of tissues and other contexts offer a potentially powerful means to address questions about different mechanisms of plastic gene regulation with respect to expression contexts [ 84 , 85 ]. In addition, it remains unresolved how often it is the case that observed context-dependent expression is, in fact, adaptive.…”

“…Mammalian genomes are diploid and consist of 2 parental copies for each gene locus [ 1 ]. Allele-specific expression (ASE) occurs in diploid genomes when one allele is preferentially expressed over the other [ 2 , 3 ]. Genetic and epigenetic differences between alleles are frequently associated with ASEs occurring at different developmental stages and leading to functional consequences for a variety of biological processes [ 2 , 3 ].…”
