To evaluate the genetic diversity of a mangrove species and clarify the genetic structure of its populations, we studied nucleotide polymorphism in two DNA regions of Bruguiera gymnorhiza collected from the southern islands of Japan, Thailand, Malaysia, Indonesia, Micronesia, and India. The two DNA sequences were the chloroplast (cp) intergenic spacer between trnL and trnF genes (ca. 300 bp), and a part (ca. 550 bp) of the nuclear gene coding for glyceraldehyde-3-phosphate dehydrogenase (GapCp). Little polymorphism was found within each of the three geographical regions, Pacific Ocean, Bay of Bengal and Arabian Sea. Throughout the vast regions east of the Malay peninsula including Indonesia, Thailand, Micronesia and the southern islands of Japan (Pacific Ocean), essentially only one haplotype (apart from variation in number of a T repeat) was present. A second haplotype was present on the western coast of Malay Peninsula and the eastern coast of India (Bay of Bengal). On the southwest of Malay Peninsula both of these haplotypes were present. Finally a third haplotype was found only on the western coast of India (Arabian Sea). When taken over all geographic populations, total nucleotide variation within the species was large (l = 0.006, average of the two genes). Our results are consistent with the hypothesis that this low genetic diversity within any local population and differentiation between the different oceans or regions are caused by very low gene flow between each of the different oceans coupled with frequent fluctuation of population sizes due to the change in sea level. The significance of these results is discussed from evolutionary point of the mangrove forests.

Attempts were made to quantify the carbon and nitrogen pools in a monospecific and pioneer mangrove stand of Kandelia obovata Sheue, Liu & Yong, Okinawa Island, Japan. The leaf C and N concentrations on a leaf area basis decreased with increasing PPFD (Photosysthetic Photon Flux Density). The total C and N stocks in foliage were estimated as 3.55 Mg ha -1 and 0.105 Mg ha -1 , respectively. The bark (45.6-48.6% for C and 0.564-0.842% for N) contained significantly higher amount of C (P < 0.05) and N (P < 0.01) than wood (46.2-47.8% for C and 0.347-0.914% N). The total C stock of stem was 23.2 Mg ha -1 in wood and 8.33 Mg ha -1 in bark, and the total N stock was 0.222 Mg ha -1 in wood and 0.116 Mg ha -1 in bark. The root wood (37.1-45.0%) contained significantly higher amount of C than root bark (35.4-40.7%) (P < 0.01). The total C stock of root was 14.2 Mg ha -1 in wood and 12.6 Mg ha -1 in bark, and the total N stock of root was 0.157 Mg ha -1 in wood and 0.155 Mg ha -1 in bark. The soil organic C and total N stocks within 1 m soil depth were estimated as 57.3 Mg ha -1 and 2.73 Mg ha -1 , respectively. The C pool in aboveground biomass (35.1 Mg ha -1 ) was 1.3 times as large as that in belowground biomass (26.9 Mg ha -1 ). However, the soil organic C pool (57.3 Mg ha -1 ) was similar to the total C pool (62.0 Mg ha -1 ) of vegetation, indicating that the mangrove stored a large part of production in the soil. About 50% of the C was in the soil. The N pool in aboveground biomass (0.442 Mg ha -1 ) was 1.4 times as large as that in belowground biomass (0.312 Mg ha -1 ). The soil N stock was 3.3 times as large as the biomass N stock (0.754 Mg ha -1 ).

Mangrove tree species form ecologically and economically important forests along the tropical and subtropical coastlines of the world. Although low intrapopulation genetic diversity and high interpopulation genetic differentiation have been detected in most mangrove tree species, no direct investigation of pollen and propagule dispersal through paternity and/or parentage analysis and spatial genetic structure within populations has been conducted. We surveyed the mating system, pollen and propagule dispersal, and spatial genetic structure in a natural population of Kandelia candel, one of the typical viviparous mangrove tree species, using nuclear and chloroplast microsatellite markers. High diversity and outcrossing rates were observed. Paternity and parentage analysis and modelling estimations revealed the presence of an extremely short-distance component of pollen and propagule dispersal (pollen: 15.2+/-14.9 m (SD) by paternity analysis and 34.4 m by modelling; propagule: 9.4+/-13.8 m (SD) by parentage analysis, and 18.6 m by modelling). Genetic structure was significant at short distances, and a clumped distribution of chloroplast microsatellite genotypes was seen in K. candel adults. We conclude that the K. candel population was initiated by limited propagule founders from outside by long-distance dispersal followed by limited propagule dispersal from the founders, resulting in a half-sib family structure.
