Genetic diversity of HMW glutenin subunit in Chinese common wheat (Triticum aestivum L.) landraces from Hubei province

Abstract: The high molecular weight (HMW) glutenin subunit composition of 111 common landraces of bread wheat collected from Hubei province, China has been determined by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Ninety six of the accessions were homogeneous for HMW glutenin subunit composition and 15 were heterogeneous. For the Glu-1 loci, 16 alleles were detected, 3 at the GluA1locus, 9 at the Glu-B1and 4 at the Glu-D1. Three novel alleles were identified, two at the Glu-B1 and one at the G… Show more

“…A large number of rare alleles was reported, which were confirmed by the findings of Branlard (2003), Tabasum (2011), andDessalegn (2011). Several novel HMW-GS alleles, such as 7*, 7**, 8*, and 8** (Liu et al, 2007), were detected in the present study and have also been detected in Chinese wheat landraces.…”

“…Because most wheat breeding programs used 5+10, subunits 17+18, 13+16, and 14+15 should also be used, as they have a positive influence on the grain quality indices of wheat. As a result, genetic diversity as well as end-use quality would improve (Liu et al, 2007). The exploration of diversity in available genetic stocks and quality associated with favorable alleles is important, and can be effectively utilized to develop new cultivars and improve crops.…”

“…All accessions were obtained from the Institute of Crop Sciences, Chinese Academy of Agricultural Sciences, Beijing. Chinese Spring, ZM11217, ZM11221, and 12 European bread wheat and durum cultivars from Rothamsted Research Harpenden, UK, were used as standards to identify HMW-GS alleles (Liu et al 2007). …”

“…Methods for seed protein extraction and SDS-PAGE were as described by Liu et al (2007). All accessions were initially analysed using five randomly selected seeds with an additional 20 seeds being analysed for those found to be heterogeneous.…”

“…2). After various authors (Deng et al 2005, Liu et al 2007, Rhazi et al 2009) both of the mentioned GS at the locus Glu-B1 have positive effect, while GS 4+12 has negative effect on the breadmaking quality. Bekes et al (2008) states that GS 14+15 at the locus Glu-B1 is predominant in the Swedish cultivars but rare in the Finnish ones, which suggests possible northern origin.…”
