Genetic diversity of Gracilaria changii (Gracilariaceae, Rhodophyta) from west coast, Peninsular Malaysia based on mitochondrial cox1 gene analysis

Abstract: Mitochondrial cytochrome c oxidase subunit I (cox1) was employed to investigate the intraspecific genetic diversity of Gracilaria changii collected from various localities distributed along the west coast of Peninsular Malaysia. Gracilaria changii is an agarophyte with potential for commercialization in Malaysia as it has high yields of good quality agar with high gel strength for the production of food grade agar and agarose. The phylogeographic aspect of G. changii has not been studied despite its abundance … Show more

“…Several studies assessing genetic diversity in the Gracilariaceae yielded intraspecific divergence; for example, 0.17 % in Gracilaria vermiculophylla (Yow et al 2011). Ten samples of H. edulis were genetically close (0-1.1 %), which were noticeably variable from the Japanese ones (2.0-2.6 %).…”

“…The PCR amplification was carried out in an Eppendorf Mastercycler ep gradient S thermocycler (Eppendorf, Hamburg, Germany) using the parameters as described by Yow et al (2011). All PCR products were electrophoresed through a 1% agarose gel stained with SYBR ® Safe DNA gel stain (Invitrogen, USA) to check the quality before being purified by LaboPass Gel and PCR Clean-up kit (Cosmo Genetech, South Korea).…”

“…Different molecular markers have been employed in studies of phylogeny and genetic diversity of Gracilariales; these include mitochondrial markers, like COI (Yang et al 2008;Yow et al 2011Yow et al , 2013 and the cox2-cox3 spacer (Rueness 2005;Destombe et al 2010;Pareek et al 2010;Garcı´a-Rodrı´guez et al 2013;Yang et al 2013); plastidencoded markers, like the RuBisCo spacer (Destombe & Douglas 1991;Byrne et al 2002;Iyer et al 2005;Guillemin et al 2008) and rbcL M.S. Kim et al 2006); and nuclear markers, like ITS (Goff et al 1994).…”
