One of the major problems of adapting the System of Rice Intensification (SRI) techniques is knowledge shortfall on the technical skills for processing quality seeds to achieve uniform seedling establishment in the nursery and quick seedling recovery in the puddled field. Therefore, this study aimed to create suitably seed sorting technique leading to vigorous and quality seedlings to improve potentiality of SRI nursery and reduce seedlings transplanting shock. It involved sorting of MR219 rice seeds the most popular grown variety in Malaysia in NaCl solutions of 0 g/L(water alone), 40 g/L, 60 g/L, 80 g/L, 100 g/L and 120 g/L, respectively. The experiment revealed that 100% germination after ten days was obtained from the sunken MR219 seeds collected in 80 g/L of NaCl solution. These values reported a decrease in germination (85%) with increasing NaCl concentration (120 g/L), with least germination vigor of 42% in water alone (0 g/L) 3 days. The percentage of sprouting proved to be high from the sunken seeds obtained in 80 g/L with 100% sprouting success rate. A decrease in percentage (70%) has been revealed with increasing NaCl concentration from the seeds obtained in 120 g/L and also when it was reduced to 40 g/L which reported 65% of the sprouting rate. Therefore, this technical information serves as benchmark to practicing farmers that high concentration in NaCl not only reduces the percentage of viable seeds but also increases seedling preparation cost as well as the entire production cost.

Abstract:The status of rice as a model crop and the sequencing of the indica and japonica genomes have provided breeders with the necessary tools for marker assisted breeding. Simple Sequence Repeat (SSR) markers are easily available for any region of the genome, and candidate gene markers are being developed rapidly. The likely targets of MAS include yield and agronomic traits, cooking and nutritional quality, and resistances to abiotic and biotic stresses. MAS for gene pyramiding for disease and insect resistances is being widely used. For major genes and QTLs of larger effect, marker assisted backcrossing (MAB) is an effective method for developing improved versions of widely-grown "mega" varieties. Developing submergence tolerant mega varieties is a good example of how the MAB approach can result in significantly improved mega varieties within two to three years. The use of markers in more conventional breeding nurseries has been limited by cost, but is beginning to be applied for some traits like grain quality. Lower-cost marker methods combined with large-scale gene discovery will increase the use of MAS over the next decade.

Seed component structures were grouped into maternal and offspring (embryo and endosperm) tissues to characterize a dormancy quantitative trait locus (QTL) for tissue-specific function using a marker-assisted genetic approach. The approach was devised to test if genotypic/allelic frequencies of a marker tightly linked to the QTL deviate from Mendelian expectations in germinated and nongerminated subpopulations derived from a segregation population of partially after-ripened seeds and was applied to the dormancy QTL qSD12 and qSD7-1 in a nearly isogenic background of rice. Experimental results unambiguously demonstrated that qSD12 functions in the offspring tissue(s) and suggested that qSD7-1 may control dormancy through the maternal tissues. These experiments also provide the first solid evidence that an offspring tissueimposed dormancy gene contributes to the segregation distortion in a mapping population developed from partially after-ripened seeds and, in part, to the germination heterogeneity of seeds from hybrid plants. Offspring and maternal tissue-imposed dormancy genes express in very early and late stages of the life cycle, respectively, and interact to provide the species with complementary adaptation strategies. The qSD12 locus was narrowed to the region of $600 kbp on a high-resolution map to facilitate cloning and marker-assisted selection of the major dormancy gene.
