Genetic Disruption of 21-Hydroxylase in Zebrafish Causes Interrenal Hyperplasia

Abstract: Congenital adrenal hyperplasia is a group of common inherited disorders leading to glucocorticoid deficiency. Most cases are caused by 21-hydroxylase deficiency (21OHD). The systemic consequences of imbalanced steroid hormone biosynthesis due to severe 21OHD remains poorly understood. Therefore, we developed a zebrafish model for 21OHD, which focuses on the impairment of glucocorticoid biosynthesis. A single 21-hydroxylase gene (cyp21a2) is annotated in the zebrafish genome based on sequence homology. Our in s… Show more

“…Cytochrome P450 family 17 polypeptide 2 (cyp17a2), created as previously described (Eachus et al, 2017), both prolyl hydroxylase 3 (phd3; BC066699), and lactate dehydrogenase A (ldha1; BC067188) probes, generated as previously described (van Rooijen et al, 2009;Santhakumar et al, 2012).…”

“…Previous work also suggested that HIF promotes POMC activity in the mouse hypothalamic region(Zhang et al, 2011). On the other hand, Cyp17a2 is an enzyme involved in steroid hormone biosynthesis at the level of interrenal gland, which is activated upon ACTH stimulation(Ramamoorthy and Cidlowski, 2016;Eachus et al, 2017;Weger et al, 2018).…”

“…The short-term exposure to cortisol also induced changes in the expression of glucocorticoid responsive gene (fkbp5) and glucocorticoid receptors (grα and grβ) in the zebrafish testicular explants. FK506 binding protein 5 (Fkbp5) is a glucocorticoid receptor chaperone protein, also known to be a robust glucocorticoid responsive gene in zebrafish [41,53,58]. This observation was confirmed in Fdx1b-deficient zebrafish, where males showed a decreased expression of fkbp5 in response to the lower plasma cortisol levels [53].…”

“…Indeed, recent studies have demonstrated that cyp11a2 , fdx1b and hsd3b1 , but not their corresponding paralogues, are involved in cortisol synthesis in zebrafish . In addition, we have recently shown a key role for cyp21a2 in glucocorticoid biosynthesis in zebrafish …”

“…Therefore, it is highly likely that cyp17a2 is involved in the biosynthesis of glucocorticoids during larval stages, whereas cyp17a1 might be important for steroidogenesis later during development, when the gonads are developed and sex steroid synthesis in fish is also required. Support for this assumption is provided by observations showing that androgen precursor androstenedione cannot be detected in 120 hpf zebrafish larvae and that both cyp17 genes can be detected in the kidney head of adult zebrafish with much higher expression levels for cyp17a1 (28 fpkm) than for cyp17a2 (0.3 fpkm) (ArrayExpress database; http://www.ebi.ac.uk/arrayexpress, accession number E‐MTAB‐460).…”
