Previous studies have demonstrated multiple herbicide resistance in California populations of Echinochloa phyllopogon, a noxious weed in rice (Oryza sativa) fields. It was suggested that the resistance to two classes of acetolactate synthase-inhibiting herbicides, bensulfuron-methyl (BSM) and penoxsulam (PX), may be caused by enhanced activities of herbicide-metabolizing cytochrome P450. We investigated BSM metabolism in the resistant (R) and susceptible (S) lines of E. phyllopogon, which were originally collected from different areas in California. R plants metabolized BSM through O-demethylation more rapidly than S plants. Based on available information about BSM tolerance in rice, we isolated and analyzed P450 genes of the CYP81A subfamily in E. phyllopogon. Two genes, CYP81A12 and CYP81A21, were more actively transcribed in R plants compared with S plants. Transgenic Arabidopsis (Arabidopsis thaliana) expressing either of the two genes survived in media containing BSM or PX at levels at which the wild type stopped growing. Segregation of resistances in the F2 generation from crosses of R and S plants suggested that the resistance to BSM and PX were each under the control of a single regulatory element. In F6 recombinant inbred lines, BSM and PX resistances cosegregated with increased transcript levels of CYP81A12 and CYP81A21. Heterologously produced CYP81A12 and CYP81A21 proteins in yeast (Saccharomyces cerevisiae) metabolized BSM through O-demethylation. Our results suggest that overexpression of the two P450 genes confers resistance to two classes of acetolactate synthase inhibitors to E. phyllopogon. The overexpression of the two genes could be regulated simultaneously by a single trans-acting element in the R line of E. phyllopogon.

Horseweed, [Conyza canadensis (L.) Cronq.], has been the most frequent weed species to develop resistance to glyphosate in various parts of the world, including Greece. In order to investigate the resistance mechanism, susceptible (S) and resistant ®) populations collected from regions across Greece were studied. Real-time PCR was used to determine the expression levels of the key enzyme EPSPS and the four ABC transporter genes M10, M11, M7 and P3. The expression level of those genes was studied at early (1DAT) or late (4DAT) times after glyphosate treatment, applied at normal (720 g a.i. ha −1 ) and high (5.760 g a.i. ha −1 ) glyphosate rate. The proposed resistance mechanism was found not to be due to a point mutation at codon 106 of the EPSPS gene that regulates glyphosate metabolism (target-site resistance), but rather involved synchronization of the overexpression of EPSPS and ABC-transporter genes. This synchronization mechanism was based on (1) the time of induction and duration of gene overexpression, and (2) regulation by the initial glyphosate load.

The understanding of mechanism of herbicide resistance in weeds is essential for adequate or innovative weed management practices. The aim of this study was to identify and analyze the expression of genes related to degradation enhancement of imazethapyr in barnyardgrass (Echinochloa crus-galli L. Beauv.). One susceptible (SUSSP01) and two populations previouslly identified as resistant to imazethapyr (ARRGR01 and PALMS01) were used. Gene expression of CYP and GST, the translation initiating factor eIF4B, and ALS genes were evaluated after imazethapyr spraying. A reference gene stability analysis was carried out, wherein the genes 18S and actin showed to be more stable in response to the population and herbicide treatment. The gene expression analysis was performed by qRT-PCR. There was no difference in the relative expression of the ALS gene. The CYP81A6 and GSTF1 genes showed higher relative expression in the resistant populations. The CYP81A6 gene had expression 9.61 and 8.44 higher in the resistant populations ARRGR01 and PALMS01, respectively, in comparison with the untreated susceptible population. The expression of this gene was induced by spraying the herbicide imazethapyr. The GSTF1 gene showed higher relative expression in PALMS01 population, reaching 12.30 times higher in plants treated with imazethapyr in relation to untreated susceptible population. The expression of eIF4B gene in the resistant populations treated with imazethapyr was about six times higher than observed in susceptible population. The high relative expression of CYP81A6 and GSTF1 genes indicate the importance of degradation enhancement for the resistance of barnyargrass to imazethapyr.
