Genetic Composition of Laboratory Stocks of the Self-Fertilizing Fish Kryptolebias marmoratus: A Valuable Resource for Experimental Research

Abstract: The hermaphroditic Mangrove Killifish, Kryptolebias marmoratus, is the world's only vertebrate that routinely self-fertilizes. As such, highly inbred and presumably isogenic “clonal” lineages of this androdioecious species have long been maintained in several laboratories and used in a wide variety of experiments that require genetically uniform vertebrate specimens. Here we conduct a genetic inventory of essentially all laboratory stocks of the Mangrove Killifish held worldwide. At 32 microsatellite loci, the… Show more

“…The study fish had two different selfing backgrounds: the R strain was derived originally from Belize in the early 1990s, and the DAN strain was also derived from Belize but in the early 2000s [26]. These strains were chosen due to known differences in their levels of genotypic diversity (i.e.…”

“…Kryptolebias marmoratus hermaphrodites at least 6 months of age (Slc8E strain) (Tatarenkov et al, 2010) were obtained from the breeding colony maintained in the Hagen Aqualab, University of Guelph. Fish were reared individually in 120 ml semi-transparent plastic containers (FisherBrand…”

“…This clonal line is homozygous at all loci tested by microsatellite analysis and is identical to all laboratories that culture it (Tatarenkov et al, 2010). Hermaphroditic individuals were kept individually in 1.5-L plastic aquaria in a controlled environment (25 6 1 C; 14% salinity; 12 hr:12 hr light:dark photoperiod), and were fed daily ad libitum on Artemia.…”

“…This reproductive mode provides a genetic architecture composed mainly of homozygous inbred strains, and a low percentage of highly heterozygous individuals induced by outcrossing, providing the basis for the formation of new clonal lines (Mackiewicz et al, 2006). Recently, 21 distinct clonal lines have been genetically verified (Tatarenkov et al, 2010). The Hon9 line, used in the present study, is amongst these distinct and established clonal lines.…”
