Genetic characterization of the complete genome of a mutant canine parvovirus isolated in China

Abstract: A field canine parvovirus (CPV) strain, CPV-SH14, was previously isolated from an outbreak of severe gastroenteritis in Shanghai in 2014. The complete genome of CPV-SH14 was determined by using PCR with modified primers. When compared to other CPV-2 strains, several insertions, deletions, and point mutations were identified in the 5' and 3' UTR, with key amino acid (aa) mutations (K19R, E572K in NS1 and F267Y, Y324I and T440A in VP2) also being observed in the coding regions of CPV-SH14. These results indicate… Show more

“…Moreover, most of the current new CPV-2a had alanine at amino acid position 440; however, most of the new CPV-2b still had threonine. Amino acid substitution at position 440 (Thr-Ala) was also reported in the previous studies in China [19], India [21,22], Nigeria [24,31], Pakistan [32], South Africa [33], South Korea [25], and Uruguay [26]. Amino acid position 440 is also important because it is located at the top of the three-fold spike, the main antigenic site of the virus [34,35].…”

“…The amino acid substitution at residue 324 (Tyr-Ile) was also reported in Brazil [18], China [19], Hungary [20], India [21,22], Italy [23], Nigeria [24], South Korea [25], Taiwan [1,15], and Uruguay [26]. According to the above, amino acid position 324 has been shown to undergo a strong positive selection in the parvovirus of all carnivores [27].…”

“…After its emergence, the original type CPV‐2 was replaced by three antigenic variants termed CPV‐2a, CPV‐2b and CPV‐2c (Buonavoglia et al, ; Parrish et al, ; Parrish, O’Connell, Evermann, & Carmichael, ). During the years, several amino acid (aa) changes were accounted in the VP2 gene sequence (Battilani et al, ; Geng et al, ; Jeoung, Ahn, & Kim, ; Nakamura et al, ; Truyen, ) and, only recently, the analysis of the NS1 gene sequence was included in the CPV phylogenies (Canuti, Rodrigues, Whitney, & Lang, ; Grecco et al, ; Han et al, ; Li et al, ; Mira et al, ; Pérez et al, ; Zhuang et al, ).…”

“…Loop 1, Loop 2, and Loop 4 not only constitute the spike of the triple folding ( 24 ) but also form the antigen A site with four key amino acid sites (93, 222, 224, and 426). Residue 440 is located on the top of the 3-fold spike as a main viral genetic site, which might result in the emergence of further genetic variants ( 25 ). Loop 3 not only forms the shoulder of the triple folding but also participates in the formation of the antigen B site, with the key amino acid sites 299, 300, and 302 ( 26 ).…”
