Genetic Cause of Autosomal Recessive Hereditary Nephropathy in the English Cocker Spaniel

Abstract: Background: Autosomal recessive hereditary nephropathy (ARHN) in the English Cocker Spaniel is caused by a type IV collagen defect, but the underlying mutation is unknown. Animals: One hundred thirty-four English Cocker Spaniels (12 with ARHN, 8 obligate carriers, and 114 others), 3 mixed breed dogs with X-linked hereditary nephropathy (XLHN), and 7 other dogs without hereditary nephropathy were included. Methods: Diagnosis of ARHN was based on transmission electron microscopy and immunostaining of kidney. Qua… Show more

“…The Bernese mountain dog has an increased risk for immune-mediated glomerulonephritis, the English cocker spaniel for a glomerular basement membrane disorder, the soft-coated wheaten terrier for a podocytopathy and the cairn terrier for polycystic KD (McKenna and Carpenter 1980, Reusch and others 1994, Davidson and others 2007, Littman and others 2013). Dogs of the Doberman pinscher, Dalmatian and rottweiler breeds have been suggested to be at risk for glomerulopathies (Picut and Lewis 1987, Hood and others 2002b, Wakamatsu and others 2007).…”

“…Different types of canine inherited renal diseases have been described in the Bull Terrier [ 7 ], Cocker Spaniel [ 8 , 9 , 10 , 11 , 12 ], Samoyed [ 13 , 14 ], Shih Tzu [ 15 ], Soft Coated Wheaten Terrier [ 16 , 17 ], Navasota [ 18 ], English Springer Spaniel [ 19 ] and Bernese Mountain Dog [ 20 , 21 ]. Canine inherited renal diseases mostly affecting the glomeruli have been reported in the Doberman Pinscher [ 22 , 23 ], Newfoundland [ 24 ] and Bullmastiff [ 25 ].…”

“…Most of these diseases are progressive and eventually fatal, and the primary renal defect and underlying mode of inheritance are mostly unknown. Hereditary nephritis (HN) in English Cocker Spaniels, 11 Samoyeds, 12 and mixed breed dogs from Navasota 13 is the only progressive JGN with a defined molecular genetic basis. HN refers to a group of genetic disorders in which an inherited defect of type IV collagen in glomerular basement membranes (GBM) causes progressive glomerular disease 14–16 .…”
