Genetic Associations of Autopsy-Confirmed Vascular Dementia Subtypes

Abstract: Background/Aims: Genetic risk factors have not been clearly established for vascular dementias (VaD) related to stroke and cerebrovascular disease. Methods: Samples were genotyped for APOE, MTHFR and ICAM. Aβ levels and choline acetyltransferase (ChAT) activities were assayed in controls and individuals with VaD. Results: Associations were found between the APOE-Ε4 allele and mixed dementia, infarct/stroke dementia and subcortical ischemic vascular dementia (SIVD), and higher Aβ1-42 levels and decreased ChAT a… Show more

“…CADASIL, a genetic form of SVaD, is associated with Notch3 mutations whose location may differ by geography or demography (Chabriat et al, 2009). The identification of phenotypes that can be reliably and effectively determined in large samples of subjects is a critical challenge for genetic studies of VCI (Leblanc et al, 2006; Jones et al, 2011). …”

“…Despite this association, however, Sjögren et al [16, 17] failed to observe any effect of APOE ε4 on CSF levels of NFL; these results suggest that the ε4 allele may not be implicated in subcortical axonal degeneration, despite its association with increased AD risk and, to a lower degree, with cerebrovascular disease [18]. …”

“…12,13 Compared with other types of dementia, VD has a better prognosis and can be prevented to a certain extent. It is very important to discuss the risk factors, since targeted prevention has important clinical significance.…”
