Genetic architecture of recent-onset dilated cardiomyopathy in Moravian region assessed by whole-exome sequencing and its clinical correlates

Abstract: Aims. Recent-onset dilated cardiomyopathy (RODCM) is a disease of heterogeneous aetiology and clinical outcome. In this pilot study, we aimed to assess its genetic architecture and correlate genotype with left ventricular reverse remodelling (LVRR). Patients and Methods. In this multi-centre prospective observational study, we enrolled 83 Moravian patients with RODCM and a history of symptoms of less than 6 months, for whole-exome sequencing (WES). All patients underwent 12-month clinical and echocardiographic… Show more

“…The Illumina Hiseq 2500 system at the Genomic facility in University Hospital Motol was used for DNA sequencing of the captured barcoded DNA library. The resulting FASTQ files were aligned to the Human Genome Reference (hg19) using Novoalign (3.02.10) and processed as described previously (Chaloupka et al, 2018). Mutation identified in DYNC2H1 (ENSG00000187240, ENST00000398093) was confirmed by Sanger sequencing.…”

“…Lamin A/C (LMNA) is an intermediate filament protein that forms the nuclear envelope. It is well known as being mutated in patients with a high risk of malignant arrhythmias and its mutations are also related to poor LVRR [193][194][195].…”
