Genetic and Biochemical Characterization of an Exopolysaccharide With in vitro Antitumoral Activity Produced by Lactobacillus fermentum YL-11

Abstract: In the present study, the whole genome sequence of Lactobacillus fermentum YL-11, a novel exopolysaccharide (EPS)-producing lactic acid bacteria (LAB) strain isolated from fermented milk, was determined. Genetic information and the synthetic mechanism of the EPS in L. fermentum YL-11 were identified based on bioinformatic analysis of the complete genome. The purified EPS of YL-11 mainly comprised galactose (48.0%), glucose (30.3%), mannose (11.8%), and arabinose (6.0%). In vitro, the EPS from YL-11 exhibited i… Show more

“…Nevertheless, the structure observed for LBIO1 and LBIO28 EPSs resembles that found with the same technique for ropy EPS-producing bifidobacteria [10]. The use of SEM for the visualization of some EPS-producing lactobacilli, which involves different fixation steps prior to coating, gives similar homogeneous sheet-like structures [35]. However, more open-loose (porous) structures, such as the ones detected for LBIO14 EPS, were also reported [36].…”

“…The genetic organization of eps clusters in lactobacilli was classified by Deo et al [ 17 ] into eight groups according to the presence and absence of genes through hierarchical clustering analysis. Some of these groups were termed “generic” which indicates the presence of epsABCDE genes at the 5′ end, as in the case of the well-described strain of L. fermentum YL-11 [ 36 ]; other eps clusters lacking the stretch epsABCDE , either because some of these genes are in a different position in the EPS cluster or are absent, were classified as “non-generic”. According to the sequence alignment, the organization of the eps cluster of L. fermentum D12 can be classified as “non-generic” and belonging to group 8 (Additional file 2 : Fig.…”

“…In a previous study by Wei et al (2019), it was suggested that exopolysaccharide (EPS) extracted from L. fermentum YL-11 could inhibit Caco-2 and HT-29 cell lines by up to 45.6 ± 6.1%. They proved that EPS could act as an antiproliferative agent in these two types of CRC cell lines (Wei et al, 2019).…”
