Genetic Analysis of Transfer-Related Regions of the Vancomycin Resistance Enterococcus Conjugative Plasmid pHTβ: Identification of oriT and a Putative Relaxase Gene

Abstract: The pHT plasmids pHT␣ (65.9 kbp), pHT␤ (63.7 kbp), and pHT␥ (66.5 kbp) are highly conjugative pheromone-independent pMG1-like plasmids that carry Tn1546-like transposons encoding vancomycin resistance. pHT␤ is the prototype plasmid, and the pHT␣ and pHT␥ plasmids are derivatives of the insertion into pHT␤ of an IS232-like (2.2 kbp) element and a group II intron (2.8 kbp), respectively. The complete nucleotide sequence of the pHT␤ plasmid was determined and, with the exception of the Tn1546-like insertion (10,8… Show more

“…pMG1-like plasmids were widely disseminated in vancomycin-resistant E. faecium clinical isolates obtained from a hospital in the United States (27). Recently, we also isolated pMG1-like plasmids carrying Tn1546-like transposons that encode vancomycin resistance in E. faecium clinical isolates in Japan (26,28). All of the pMG1-like plasmids carry a conserved traA gene which is involved in the tra gene system for conjugation and is pMG1 specific (24).…”

“…Broth mating and solid-surface mating were performed as previously described (48,49), with a donor/recipient ratio of 1:10. Broth matings (in THB) were carried out for 4 h, and solid-surface matings (on THB agar plates) were carried out overnight (16 h) at 37°C.…”

“…Conjugative transfer of plasmids encoding the synthesis of several products such as bacteriocins has been well studied by Rauch et al [2]. The investigation of mechanisms and conditions allowing plasmid transfer and expression of plasmid-encoded genes in host cells have been reported [3,4,5]. B. thuringiensis is the most known insecticidal bacterium worldwide for the control of various plant pests.…”
