Genetic analysis of methionine-rich storage protein accumulation in maize

Abstract: Experiments were conducted to determine the chromosomal location of the gene conditioning overproduction of a methionine-rich, 10-K zein in maize kernels of line BSSS53. In addition, the chromosomal location of the structural gene encoding the overproduced protein was determined. Whereas the structural gene, designated Zps10/(22), was found to be located on the long arm of chromosome 9 near the centromere, the locus regulating overproduction of the zein protein was mapped to the short arm of chromosome 4. This… Show more

“…Genetic improvement of such endosperm traits has received considerable attention in plant breeding (Benner et al, 1989;Sadimantara et al, 1997;Mazur et al, 1999). Quantitative genetics models for analyzing the triploid inheritance of endosperm traits have been developed and applied to practical breeding populations in cereals (Gale, 1976;Mo, 1987;Bogyo et al, 1988;Foolad and Jones, 1991;Pooni et al, 1992;Zhu and Weir, 1994;Wu et al, 1998).…”

“…However, we have previously shown that supplemented methionine can be replaced by corn-based feed with elevated levels of the 10-kDa zein (Messing and Fisher, 1991). The elevated levels are due to the presence of a trans-acting factor, dzr1, encoded by a gene on chromosome 4S in inbred BSSS53 (Benner et al, 1989). The dzr1 locus has been linked to a cluster of 22-kDa zein genes and the RFLP marker php20725 Chaudhuri and Messing, 1995).…”

“…However, dzrl, on the short arm of chromosome 4 seems to affect only the relative expression of the 10-kDa 5 zein gene (Benner et al, 1989;Chaudhuri and Messing, 1995). Moreover, we have identified different alleles of this regulator.…”

“…Variability in seed-methionine levels, however, not only provide an opportunity to study tissue-specific regulation of gene expression, but also to corrcxt a nutritional deficiency in animal feed, where methionine is the only essential amino acid that is supplemented (Messing and Fisher, 1993). We were able to show that variability of seed methionine was due to the post-transcriptional regulation of the 10-kDa zein gene (Cruz-Alvarez et al, 1991; Schickler et al, 1993) by alleIes of a trans-acting locus dzrl (Benner et al, 1989;Chaudhuri and Messing, 1995).…”
