Genetic Analysis of a Gene Cluster for Cyclohexanol Oxidation in Acinetobacter sp. Strain SE19 by In Vitro Transposition

Abstract: Biological oxidation of cyclic alcohols normally results in formation of the corresponding dicarboxylic acids, which are further metabolized and enter the central carbon metabolism in the cell. We isolated an Acinetobacter sp. from an industrial wastewater bioreactor that utilized cyclohexanol as a sole carbon source. A cosmid library was constructed from Acinetobacter sp. strain SE19, and oxidation of cyclohexanol to adipic acid was demonstrated in recombinant Escherichia coli carrying a SE19 DNA segment. A r… Show more

“…These results and those of the phylogenetic analysis suggest that the unknown protein from A. pleuropneumoniae (2) (ORF1Ap), protein ID317 from B. japonicum (21), ChnZ from Acinetobacter sp. strain SE19 (9), and the putative transposase from a Wolbachia endosymbiont of D. simulans (accession number AAK96114) might be encoded by complete or truncated ISs. These sequences, along with ISCce1, might form a new group of ISs, which is probably related to the IS481 and the IS3 families.…”

“…14,15) It is known that the bacterial metabolism of cyclohexanol to adipic acid proceeds via cyclohexanone, "-caprolactone, 6-hydroxyhexanoic acid, and 6-oxohexanoic acid. 16) Each gene encoding the enzyme involved in this pathway has been identified, 17) so that the identification of the enzyme oxidizing cyclohexane to cyclohexanol and its coding gene is essential for studies of the cycloalkane degradation pathway. It has been thought that AH catalyzes the terminal oxidation of n-alkanes.…”

“…The alicyclic hydrocarbons are a major component of petroleum, and microbial metabolism of these compounds has been reported in various microorganisms (5,7,9,11,13,18,26). Most of the microbes isolated to date that metabolize alicyclic hydrocarbons contain degradation pathways for small (C 5 , C 6 , and C 7 ) cyclic compounds (7,12,13,15).…”
