Genes for hydrogen peroxide detoxification and adaptation contribute to protection against heat shock in Xanthomonas campestris pv. campestris

Abstract: Xanthomonas campestris pv. campestris, a soil-borne plant-pathogenic bacterium, is exposed to multiple stresses in the environment and during interaction with a host plant. The roles of hydrogen peroxide (H(2) O(2) )-protective genes (katA, katG, and ahpC) and a peroxide sensor/transcription regulator (oxyR) in the viability of X. campestris pv. campestris at an elevated temperature were evaluated. The single katA and katG mutants showed moderate decreased survival after the heat treatment, while the double ka… Show more

“…In an Anabaena sp., ahpC is associated with protection against high temperatures (47°C), high salinity, toxic pesticides (carbofuran), heavy metals, and UV irradiation (38). However, in Xanthomonas campestris, ahpC performs no protective function against heat treatment (39). The expression of AhpC proteins in Acidithiobacillus ferrooxidans (40) and Shewanella putrefaciens (41) increases when they are incubated at low temperatures of 2 to 15°C.…”

“…The PCR products were visualized by 1.8% agarose gel electrophoresis. Real-time PCR experiments were performed as described previously (23). Briefly, 10 ng of cDNA was used as a template in a Kapa SYBR Fast quantitative PCR (kit containing Master Mix 2ϫ; ABI Prism) reaction in an Applied Biosystems StepOnePlus real-time PCR system.…”
