Generative modelling of the thalamo-cortical circuit mechanisms underlying the neurophysiological effects of ketamine

Abstract: Cortical recordings of task-induced oscillations following subanaesthetic ketamine administration demonstrate alterations in amplitude, including increases at high-frequencies (gamma) and reductions at low frequencies (theta, alpha). To investigate the population-level interactions underlying these changes, we implemented a thalamo-cortical model (TCM) capable of recapitulating broadband spectral responses. Compared with an existing cortex-only 4-population model, Bayesian Model Selection preferred the TCM. Th… Show more

“…Alterations in gamma have been described in mood disorders [164][165][166] and proposed as putative biomarkers of depression [167]. Our study agrees with animal and clinical studies that have reported similar increases in gamma power following acute ketamine administration at therapeutic levels [168][169][170][171][172][173]. Similarly to DBS, subanesthetic doses of ketamine, a noncompetitive N-methyl-D-aspartate (NMDA) receptor antagonist, also provides a fast antidepressant action [174], being particularly effective in anxious depression [175] and it is also helpful in relieving social anxiety disorder [176].…”

“…Interestingly, these findings all reflect changes in intrinsic connectivity that regulate or modulate inhibition locally. Within the amygdala in particular, increased excitatory drive onto inhibitory interneurons for TRD participants seems at odds with an increased state of excitability within this region; however, similar accounts of increased pyramidal-to-inhibitory interneuron drive have previously been reported (64) and are thought to reflect a link between increased pyramidal cell excitability locally and downstream effects of increased gamma power.…”

“…Finer-grained cellular, synaptic or pharmacological resolution would require more complex models ( cf . Shaw et al ., 62 , 63 Symmonds et al . 4 ) demonstrated a reduction in fronto-temporal beta coherence by bvFTD, which was recapitulated in the loss of beta-band coherence and Granger causal connectivity in the non-fluent primary progressive aphasia variant of frontotemporal dementia.…”
