Generation of representative primary virus isolates from blood plasma after isolation of HIV-1 with CD44 MicroBeads

Abstract: Infection of cell cultures with cell-free virus isolated from HIV-infected patients is notoriously difficult and results in a loss of viral variation. Here, we describe viral sequences from PBMC, U87.CD4.CCR5 and U87.CD4.CXCR4 cell cultures and compare them to those from blood plasma from 12 patients from whom virus particles were isolated using CD44 MicroBeads. In both PBMC and U87.CD4.CCR5 cultures, 66% of the plasma viral strains were retrieved after culturing. In addition, coreceptor use was predicted base… Show more

“…Among the patient plasma samples assayed, PSGL-1 was found to be present on all of the viruses tested and at similar levels to the positive control, CD44. Previous work has shown that CD44 can efficiently capture viruses from plasma of HIV-infected patients (49,50), and our results herein suggest that PSGL-1 could also be used for similar purposes in the future.…”

“…4). As an additional control, we also assayed CD44 incorporation in the same plasma samples, since CD44 is known to be incorporated with high efficiency into HIV-1 virions (49)(50)(51). CD44 was also incorporated into virions from all patient plasmas tested, with a wide range of virus input recovered by capture with anti-CD44 (8-87%), indicating that PSGL-1 falls into a category of host proteins that are incorporated with high efficiency into HIV virions.…”

“…Therefore, CD44 is likely incorporated into HIV-1 in vivo. In support of this notion, an anti-CD44 antibody and CD44 microbeads interact with viruses derived from the plasma of HIV-1 patients [22,76,97]. Interestingly, when HIV-1 is produced from CD44-expressing cells including PBMCs and hence incorporates CD44, cell-free infection of this HIV-1 is prevented by HA on CD44-expressing target CD4 + T cells at the step of virus attachment [98].…”
