Generation of Pax1/PAX1-Specific Monoclonal Antibodies

Abstract: Pax genes encode an evolutionary conserved group of transcription factors with multiple roles during embryonic development and for cell type specification in normal and malignant tissues of the adult organism. In mice, Pax1 is required for the formation of specific skeletal structures as well as for the development of a fully functional thymus. In humans, the PAX1 locus has been linked to otofaciocervical syndrome, idiopathic scoliosis, and to a higher susceptibility for androgenic alopecia. In addition, the m… Show more

“…To generate an antibody against murine Regnase-3 protein, aa 1–903 of the Regnase-3 protein sequence with N-terminal GST-tag and C-terminal His-tag were expressed in Escherichia coli and purified in two steps on a HiTrap Chelating and a GSTrap column. Lou/C rats and C57BL/6 mice were immunized with the Regnase-3 peptide as described elsewhere (Feederle et al, 2016). Specific antibodies were first selected by enzyme-linked immunoassay on the protein and, in a second step, by immunoblotting using GFP-Regnase-3 protein lysates, expressed in HEK293T cells.…”

“…To generate antibodies against human YKL-39, Lou/C rats and C57 BL/6 mice were immunized with ovalbumin-coupled peptides spanning aa 364–377 and 133–154, respectively, using standard procedures as described. 79 Specificity of antibodies was verified by enzyme-linked immunoassay on peptides and by Western blotting using stably transfected HEK293-YKL-39-FLAG and HEK293-vector cells generated by stable transfection with plasmids pSNAP-YKL-39-FLAG and pSNAP-tag. The hybridoma cells of YKL-39-reactive supernatants were cloned at least twice by limiting dilution.…”

“…Expression and purification of an N-terminal fragment of ASXL1 (amino acids 1–618), followed by immunization of Lou/c rats was performed as previously described (Feederle et al., 2016, Studier, 2005).…”
