Generation of Local CA1 γ Oscillations by Tetanic Stimulation

Abstract: Neuronal network oscillations are important features of brain activity in health and disease and can be modulated by a range of clinically used drugs. A protocol is provided to generate a model for studying CA1 γ oscillations (20 -80 Hz). These γ oscillations are stable for at least 30 min and depend upon excitatory and inhibitory synaptic activity in addition to activation of pacemaker currents. Tetanically stimulated oscillations have a number of reproducible and easily quantifiable characteristics including… Show more

“…Brain slices were prepared similar to those described previously [ 16 ]. SUS- and sham-treated mice were anaesthetized with 2% isoflurane (Attane) and transcardially perfused with cold cutting solution comprising (in mM) 125 choline-Cl, 2.5 KCl, 0.4 CaCl 2 , 6 MgCl 2 , 1.25 NaH 2 PO 4 , 26 NaHCO 3 and 20 D-glucose saturated with carbogen gas (95% O 2 /5% CO 2 ).…”

“…300 μm coronal hippocampal brain slices, from between approximately -1.34 to -2.54 mm of the Bregma, were cut on a vibratome (VT1000S, Leica). Slices were rested for 30 minutes at 35°C and then at room temperature (RT) for at least 30 minutes prior to recordings, which were conducted by an experimenter who was blinded to treatment [ 16 , 17 ].…”
