Generation and Characterization of Aggrecanase

Abstract: A method was developed for generating soluble, active "aggrecanase" in conditioned media from interleukin-1-stimulated bovine nasal cartilage cultures. Using bovine nasal cartilage conditioned media as a source of the aggrecanase enzyme, an enzymatic assay was established employing purified aggrecan monomers as a substrate and monitoring specific aggrecanase-mediated cleavage products by Western analysis using the monoclonal antibody, BC-3 (which recognizes the new N terminus, ARGS, on fragments produced by cl… Show more

“…Although it has been widely accepted (22,23,30,(37)(38)(39)(40)(41)(42) that ADAMTS4 (aggrecanase-1) cleaves only at the Glu 373 -Ala 374 bond within the IGD, the results described here suggest that both of the major cleavages that are known to occur in vivo, at Glu 373 -Ala 374 and Asn 341 -Phe 342 , can be catalyzed by ADAMTS4. Previously, cleavage at the Asn 341 -Phe 342 site in the aggrecan IGD was considered a specific property of the MMP family, with the exception of a snake venom enzyme (30,34) 341 -Phe 342 bond in the experiments described here was due to contamination of the ADAMTS4 preparation with an MMP-like activity was eliminated by the observation that although 10 mM EDTA and TIMP-3 totally blocked the cleavage, neither TIMP-1 nor TIMP-2 was inhibitory, even at 750 nM.…”

“…TIMPs have also recently been shown to inhibit metalloproteinases other than MMPs. An IC 50 value of 110 nM (0.11 M) has been reported for TIMP-3 inhibition of TACE (32), and an IC 50 value of 210 nM has been estimated for TIMP-1 inhibition of aggrecanase (13). Both TACE and aggrecanase are recently identified ADAM family members.…”

“…Determination of Aggrecan Cleaving Activity-Aggrecanase activity was measured by incubating bovine aggrecanase or recombinant human ADAMTS11 with purified bovine aggrecan monomers as described (27). Reactions were terminated with 20 mM EDTA and Western blotted.…”

“…Reactions were terminated with 20 mM EDTA and Western blotted. Glu 373 -Ala 374 cleavage products were detected using the BC33monoclonal antibody (28) IC 50 values were determined for the inhibitors using conditioned media from stimulated bovine nasal cartilage explant cultures and recombinant human ADAMTS11 as described previously (27).…”
