Gene replacement in Dictyostelium: generation of myosin null mutants.

Abstract: The eukaryotic slime mold Dictyostelium discoideum has a single conventional myosin heavy chain gene (mhcA). The elimination of the mhcA gene was achieved by homologous recombination. Two gene replacement plasmids were constructed, each carrying the G418 resistance gene as a selective marker and flanked by either 0.7 kb of 5′ coding sequence and 0.9 kb of 3′ coding sequence or 1.5 kb of 5′ flanking sequence and 1.1 kb of 3′ flanking sequence. Myosin null mutants (mhcA‐ cells) were obtained after transformation… Show more

“…discoideum wild-type and mutant cells were cultivated in petri dishes using liquid AX2 nutrient medium (provided by the Max-Planck-Institut fü r Biochemie, Martinsried, Germany). The wild-type AX2 and the myosin null mutant HS2205 [29] were transfected to express fluorescent GFP-atubulin (cell lines HG1668 and HG1671 [30,31]). For each experiment, the cells were taken from axenic culture.…”

“…For the developmental study, cells were washed twice in 12 mM Na/K phosphate buffer and resuspended at 7 ϫ 10 7 cells/ml and plated at various densities on agar plates. Manstein et al (1989). Because there are two copies of SPKA genes, spkA null cells were generated in JH10 cells by using two targeting constructs containing a Thy cassette and a Bsr cassette, respectively.…”

“…The spkA null mutant was obtained by a gene replacement technique based on that of Manstein et al (1989). Because there are two copies of SPKA genes, spkA null cells were generated in JH10 cells by using two targeting constructs containing a Thy cassette and a Bsr cassette, respectively.…”

“…Dictyostelium amoebae are haploid, and many genes, including calmodulin (Goldhagen and Clarke, 1986), that are present in multiple copies in mammalian cells are single-copy in Dictyostelium. Thus, for several Dictyostelium proteins, it has been possible to manipulate gene expression by deleting or altering the appropriate genomic sequences via homologous recombination (DeLozanne and Spudich, 1987;Witke et al, 1987;Manstein et al, 1989;Jung and Hammer, 1990;Doring et al, 1991;Kumagai et al, 1991;Sun and Devreotes, 1991;Cox et al, 1992). Reduction of mRNA levels by the introduction of vectors producing antisense RNA also has proved effective.…”
