Gene expression profile of esophageal cancer in North East India by cDNA microarray analysis

Abstract: AIM:To identify alterations in genes and molecular functional pathways in esophageal cancer in a high incidence region of India where there is a widespread use of tobacco and betel quid with fermented areca nuts. METHODS:Total RNA was isolated from tumor and matched normal tissue of 16 patients with esophageal squamous cell carcinoma. Pooled tumor tissue RNA was labeled with Cy3-dUTP and pooled normal tissue RNA was labeled with Cy5-dUTP by direct labeling method. The labeled probes were hybridized with human … Show more

“…Therefore, it comes as no surprise that deregulation of these signaling pathways frequently occurs in human cancer, including esophageal squamous cell carcinoma (Chattopadhyay et al, 2007;Li et al, 2007). Our results demonstrate that PGE 2 markedly increased the phosphorylation of Erk1/2, but not JNK or p38, in cultured esophageal squamous cell carcinoma cells (Fig.…”

“…North East India has the highest incidence of esophageal cancer and the number of esophageal cancer cases in this region is increasing at an alarming rate (Chattopadhyay et al, 2007;Sharma et al, 2013;Sharma et al, 2014b). Comparison of esophageal cancer incidence rate in the year 2006-2008 and 2009-2011 revealed a noticeable increase in the incidence rate of esophageal cancer among the North Eastern states of India (NCRP 2006(NCRP -2008(NCRP , 2009(NCRP -2011.…”

“…We also found several genes involved in zinc homeostasis including ZNF12, ZNF750 and ZNF185 were downregulated, which support the finding of earlier studies that deregulation of zinc homeostasis is associated with ESCC. 17,18 Interestingly, we found few downregulated genes in ESCC which includes ZNF185, TMPRSS11E, SPINK5, SERPINB4, SCEL, RHCG, PPP1R3C, IVL, HOP, GPX3 and PPL. These genes were also reported as downregulated in a study on esophageal adenocarcinoma using DNA microarrays.…”

“…Microarrays containing cDNA probes have been widely used to perform gene expression analysis of ESCC. 12,17,18 cDNA probes are largely being replaced by oligonucleotide probes which are commercially available and easily affordable. A study by Uchikado et al, identified 43 upregulated genes in ESCC using oligonucleotide arrays containing 17,086 probes.…”
