Gene expression analysis of wounding‐induced root‐to‐shoot communication in Arabidopsis thaliana

Abstract: Root-to-shoot communication plays an important role in the adaptation to environmental stress. In this study, we established a model system for root-to-shoot signalling to observe global gene expression in Arabidopsis thaliana. The roots of Arabidopsis seedlings were wounded and the expression in the shoots of 68 and 5 genes was up-regulated threefold at 30 min and 6 h post-injury, respectively. These genes were designated early and late Root-to-Shoot responsive (RtS) genes, respectively. Many of the early RtS… Show more

“…For example, wounding or systemin treatment of tomato roots caused induction of systemic responses in leaves and this was dependent on JA signaling (Li et al, 2002), and wounding of Arabidopsis roots caused increased JA biosynthesis in shoots but not roots (Hasegawa et al, 2011). Thus it is possible that LBD20 may be involved in the development of regulatory signals that move from roots to shoots.…”

“…In addition, ethylene (ET)-related genes such as 1-aminocyclopropane-1-carboxylic acid (ACC) synthase 6 (ACS6 ) were also induced in the shoots with wounded roots. 6,8 In fact, root-to-shoot response of some early RtS genes examined in the response. For this purpose, we introduced some RtS promoter::firefly Luc fusion genes into Arabidopsis plants.…”

“…Therefore, we tried to establish a model system to monitor the gene expression during wound-induced root-to-shoot communication in Arabidopsis seedlings. 6 Then, we have performed the transcriptome analysis of wound-induced root-toshoot response in Arabidopsis. 6 When the roots of Arabidopsis seedlings were wounded, the expression in the shoots of many genes was upregulated more than three-fold at 30 min and 6 h post-injury.…”
