Gene Expression Analysis of Ex Vivo Expanded and Freshly Isolated NK Cells From Cancer Patients

Abstract: Summary The infusion of natural killer (NK) cells is a promising therapy for patients with advanced malignancies. Clinical expanded NK cell products were compared to freshly isolated NK cells. Autologous peripheral blood mononuclear cells were collected by apheresis from 8 patients. NK cells were isolated by anti-CD3 negative selection followed by anti-CD56 positive selection. They were then expanded by coculture with IL-2 and an irradiated EBV-transformed lymphoblastoid cell line (EBV-TM-LCL) to produce 14 NK… Show more

“…Indeed, gene expression profiles were shown to undergo substantial changes during NK-cell in vitro expansion. 51 However, comparative functional experiments with non-expanded versus expanded NK cells from some healthy donors and CML patients confirmed the results obtained with expanded NK cells and argue against a major effect of in vitro expansion on NK-cell functionality.…”

“…No significant correlation of CD107a secretion with disease response at the time of analysis was found (Figure 4c). In contrast to the reduced degranulation responses, direct cytolysis of K562 by 51 Cr release was unaffected at various effector to target ratios in patients on imatinib treatment (Figure 4d, Supplementary Figure 2). Again, presence or absence of 1 mM imatinib in the culture medium during NK-cell expansion did not affect results ( Figure 4d, center panel), and non-expanded NK cells from healthy donors and patients were also equally effective (Figure 4d, right panel).…”

“…The adoptive infusion of NK cells is a promising immunotherapy for patients with advanced malignancies [5]. Using gene and microRNA expression microarrays, Park et al provided distinct expression profiles of ex vivo expanded NK cells and freshly isolated NK cells from cancer patients [17]. Among approximately 25 100 genes evaluated, the expanded NK cells overexpressed 1098 genes and 28 microRNAs when compared with freshly isolated human NK cells [17].…”

“…Indeed, EBV-LCL stimulation has been reported as a method to expand and culture NK cells for both experimental and clinical use [347][348][349][350]. In contrast, the final drug product generated cell drug product; which, has been shown to be preferred for ACT immunotherapy [353][354][355].…”
