Gender‐Specific Effects on Verapamil Pharmacokinetics and Pharmacodynamics in Humans

Abstract: Pharmacokinetic studies of i.v. and oral racemic verapamil and 14C-erythromycin breath tests (ERBT) were performed in 84 healthy men (n = 42) and women (n = 42). Verapamil was measured by HPLC, concentration versus time data were analyzed by noncompartmental models, protein binding was measured by equilibrium dialysis, and statistical analyses were performed by ANOVA. Clearance of i.v. and p.o. verapamil was 13.7 +/- 4.3 and 58.4 +/- 35 ml/min/kg (mean +/- SD) in women compared to 12.6 +/- 3.4 and 82.6 +/- 70 … Show more

“…The increased bioavailability and decreased clearance of oral verapamil in women compared to men caused a greater reduction in blood pressure and heart rate in women compared to men [ 44 ]. However, another study that examined sex-based pharmacokinetic differences for oral verapamil showed that the reduction in mean arterial blood pressure owing to the drug was closely correlated with its plasma concentration, although without any interaction by sex [ 28 ].…”

“…Sasaki et al [7] did not find any gender related differences in the pharmacokinetics of verapamil following 80 mg oral dosing with the drug. Krecic-Shepard et al reported that verapamil oral clearance was faster in men compared with women when administered as either a sustained release or a regular release formulation [8,9]. Their findings were in contrast to their previous reports of faster clearance of intravenously administered verapamil in women [6].…”

“…[8][9][10][11] Matsuo et al demonstrated that there is a correlation between the CL tot of cyclosporine and the grade severity of liver disease by Child-Pugh classification in patients with hepatitis. 12) However, it is not easy to utilize these approaches to determine the degree of residual hepatic metabolizing ability in patients, as is also the case with creatinine clearance in patients with renal failure.…”
