Gemcitabine-based chemotherapy in colon squamous cell carcinoma: A case report and literature review

Abstract: Abstract. Squamous cell carcinoma (SCC) originating from the colon is rare. In terms of its clinicopathological characteristics, this type of cancer has been reported to be more aggressive and have a worse prognosis compared with adenocarcinoma. We herein present a successful therapeutic approach applying neoadjuvant and adjuvant gemcitabine-based chemotherapy in a patient with colon SCC. A 58-year-old male patient received two cycles of neoadjuvant chemotherapy with a regimen including gemcitabine, oxaliplati… Show more

“…Gemcitabine is a nucleoside antimetabolite that inhibits DNA synthesis ( 1 ). It is most commonly used in organ malignancies due to its functions in the promotion of cell death in several cancers including non-small cell lung cancer, colon squamous cell carcinoma, nasopharyngeal carcinoma and ovarian, breast and pancreatic cancer ( 2 5 ). Gemcitabine-induced antitumor therapy resistance is a widely used model in studying chemotherapeutic resistance ( 6 ).…”

“…Chemotherapy could also be employed as a palliative treatment. Many authors agree to various treatment regimens including 5 fluorouracil (5FU)/mitomycin C, 5FU, 5FU/cisplatin, capecitabine/cisplatin, capecitabine and raltitrexed/oxaliplatin [ 3 , 4 ]. Zhao et al .…”

“…Usually, the disease progress through metastasis within the first two years of treatment and is associated with an overall poor prognosis [ 2 ]. Surgical resection is the main therapeutic modality, while the role of chemotherapy and radiotherapy remain ambiguous [ 3 ]. We report a case of SCC of the cervix metastasizing to the caecum and resulting in acute bowel obstruction.…”

“…By virtue of its distinctive features of cellular pharmacology, metabolic properties, and mechanistic characteristics, gemcitabine occupies an important place at the forefront of chemotherapy for the treatment of a wide variety of tumors (Ciccolini, Serdjebi, Peters, & Giovannetti, 2016;Crin o & Cappuzzo, 2002;Dubey, Saneja, Gupta, & Gupta, 2016;Ducoulombier, Cousin, Kotecki, & Penel, 2016;Hansen, 2001;Liu, Zhang, Li, Zhang, & Lu, 2014;Ottaiano et al, 2017;Rizzuto, Ghazaly, & Peters, 2017;Schlack, Boegemann, Steinestel, Schrader, & Krabbe, 2016;Thigpen, 2002;Zhang et al, 2017;Zhao, Guo, Sun, Lv, & Qiu, 2017). The source of cytotoxic activity of gemcitabine is primarily attributed to its ability to transport across the cell membrane to get phosphorylated efficiently, slower rate of elimination along with selfpotentiation mechanisms to mask DNA chain termination and extensive inhibitory efficiency against several enzymes (Barton- Burke, 1999;Plunkett et al, 1995;Plunkett, Huang, Searcy, & Gandhi, 1996;Plunkett, Huang, & Gandhi, 1997).…”
