Gelidium elegans Extract Ameliorates Type 2 Diabetes via Regulation of MAPK and PI3K/Akt Signaling

Abstract: Gelidium elegans, a red alga native to the Asia Pacific region, contains biologically active polyphenols. We conducted a molecular biological study of the anti-diabetic effect of Gelidium elegans extract (GEE) in C57BL/KsJ-db/db mice. Mice that had been administered GEE had significantly lower body mass, water consumption, and fasting blood glucose than db/db controls. Moreover, hemoglobin A1c (HbA1c), an indicator of the glycemic status of people with diabetes, was significantly lower in mice that had been ad… Show more

“…IRS-1 plays a key role in insulin signal transduction, and GLUT4 is necessary for insulin-stimulated glucose uptake [ 16 ]. Data suggest that the activation of MAPK signalling pathway may suppress the phosphorylation of IRS and downregulate GLUT4 expression, resulting in reduced glucose transport [ 54 ]. In the present study, resveratrol and 17β-estradiol could down-regulate p-ERK expression to inhibit the activation of MAPK, and then up-regulate GLUT4 and IRS-1 expressions, therefore promoted the glucose uptake and exerted the hypoglycemic effect.…”

“…Recently, ginsenosides Rb1, Rb2, Rg1, Rg3, Rh2, and compound K, major pharmacological components of RGE, have been reported to suppress the hepatic gluconeogenesis via adenosine monophosphate (AMP)-activated protein kinase (AMPK) [ 22 , 23 , 24 ]. Importantly, enhanced GLUT4 expression mediated by activated insulin receptor substrate (IRS)/phosphatidylinositol-3,4,5-triphosphate (PI3K)/serine-threonine protein kinase signaling pathway by the treatment of RGE or ginsenosides increased glucose uptake in adipocytes or skeletal muscle cells and, thereby, decreased blood glucose or Hb-A1c levels [ 22 , 25 ]. In addition, enhancement of glucose uptake via GLUT4 through the upregulation of adipocytic peroxisome proliferator-activated receptor-γ also contributes to the ginsenoside-mediated glucose control mechanism [ 26 ].…”

“…Another seaweed improving glucose uptake is Gelidium amansii. It exhibits antihyperglycemic, antioxidant, and antiobesity effects potentially via PI3K/AKT/GLUT4 signaling [66]. Also, carnosol, a compound found in spices such as sage or rosemary, increases glucose uptake via GLUT4 [67].…”
