GDI1 encodes a GDP dissociation inhibitor that plays an essential role in the yeast secretory pathway.

Abstract: GTP binding proteins of the Sec4/Ypt/rab family regulate distinct vesicular traffic events in eukaryotic cells. We have cloned GDI1, an essential homolog of bovine rab GDI (GDP dissociation inhibitor) from the yeast Saccharomyces cerevisiae. Analogous to the bovine protein, purified Gdi1p slows the dissociation of GDP from Sec4p and releases the GDP‐bound form from yeast membranes. Depletion of Gdi1p in vivo leads to loss of the soluble pool of Sec4p and inhibition of protein transport at multiple stages of th… Show more

“…Our data support the view that there are at least three distinct GDI isoforms (Janoueix-Lerosey et al, 1995;Pfeffer et al, 1995). Yeast (Saccharomyces cerevisiae) encodes only one GDI (Gdi1p), and it has been shown to be an essential gene product (Garrett et al, 1994). A variety of explanations have been put forward for the existence of multiple mammalian isoforms.…”

“…The binding of Rab proteins to the universal regulatory protein GDI is of crucial importance for Rab membrane cycling and thus for the regulation of vesicular trafficking (19). In order to exert their function, Rab proteins need to be recruited specifically to the cytosolic surface of intracellular membranes in a temporally controlled manner.…”

“…The Sec4 GTPase can cycle on and off membranes in a manner that is dependent on the nucleotide state of the Rab and its interaction with GDI (17). To determine whether the absence of clustering seen with the sec4-8 secretory vesicles correlated with a loss of Sec4 function and not with an indirect defect in the sec4-8 mutation on the functionality of the vesicles, we made use of purified recombinant Rab GDI to extract Rab GTPases from the vesicles.…”
