GD3, a prominent ganglioside of human melanoma. Detection and characterisation by mouse monoclonal antibody.

Abstract: We have previously described (1) a mouse IgG3 monoclonal antibody (AbR24) with a high degree of serological specificity for cultured human melanoma cells. All melanoma cell lines and two astrocytomas examined were positive for the heat-stable cell surface antigen detected by this antibody. Although choroidal melanocytes and brain had low levels of the antigen, a wide variety of other cells and tissues were unreactive. Three other monoclonal antibodies (Abs C5, I24, and K9), having a similar restricted specific… Show more

“…GD3 was also found to be expressed in malignant melanocytes, namely melanoma tumors (Portoukalian et al, 1979;Carubia et al, 1984), and has been considered to be a human melanoma marker (Pukel et al, 1982). In fact, GD3 is expressed in majority of melanoma cell lines and melanoma tumors, whereas it can be scarcely detected in normal melanocytes (Furukawa et al, 1990).…”

“…The remarkable diversity of cell-surface glycoconjugates and their regulation with cellular differentiation have been increasingly appreciated (11) . Although once considered primarily neural antigens, gangliosides are present on thymic epithelium, red blood cells, pancreatic islets, kidney glomeruli, and malignancies, including melanoma and neuroblastoma, and as shown in this study of human T lymphocytes (12)(13)(14)(15). The presence of gangliosides on the surface of these diverse cell types suggest a common though currently unknown function .…”

“…10 Anti-sLe x antibody CSLEX-1 (American Type Culture Collection, Manassas, VA), anti-Le x antibody (BD Pharmingen, Heidelberg, Germany), negative control IgM monoclonal antibody (mAb) (BD Pharmingen), antiGQ1b/GD3 mAb R24, 11 anti-PSA mAb 735, 12 anti-Le a antibody (Acris Antibodies, Hiddenhausen, Germany), and anti-GP1b mAb AK2 (Serotec, Düsseldorf, Germany) were used at 10 g/mL. Biotinylated Aleuria aurantia lectin (AAL), concanavalin A, peanut agglutinin, Sambucus nigra lectin (SNA), and Maackia amurensis lectin II (MAL II) (Vector Laboratories, Burlingame, CA) were used at 2 to 5 g/mL and detected with phycoerythrin-conjugated streptavidin (Jackson Immunoresearch, West Grove, PA).…”
