GC‐MS structural characterization of fatty acids from marine aerobic anoxygenic phototrophic bacteria

Abstract: The FA composition of 12 strains of marine aerobic anoxygenic phototrophic bacteria belonging to the genera Erythrobacter, Roseobacter, and Citromicrobium was investigated. GC-MS analyses of different types of derivatives were performed to determine the structures of the main FA present in these organisms. All the analyzed strains contained the relatively rare 11-methyloctadec-12-enoic acid, and three contained 12-methyl-octadec-11-enoic acid, which has apparently never been reported before. High amounts of th… Show more

“…The samples were shipped in a frozen state and analyzed in the Třeboň laboratory, typically within 1 mo after sample collection. As tested with Roseobacter-like strain COL2P (Rontani et al 2005), the freeze-thaw cycle of formaldehyde-fixed samples does not significantly reduce their IR emission. Chlorophyll a (chl a) content was determined by high performance liquid chromatography as described previously or spectrophotometrically in 90% acetone extracts.…”

“…The initial claim that the AAP community is dominated by Erythrobacter sp. (Kolber et al 2001) was later dismissed by both culture-free (Béjà et al 2002) and cultivation studies (Rontani et al 2005, Koblí=ek et al 2006. Rontani et al (2005) isolated 3 strains of Erythrobacter and Roseobacter species from the Baltic Sea, which confirms the presence of these genera in the environment.…”

“…strain NAP1, and Citromicrobium sp. strain CV44, were described previously (29)(30)(31)(32). Freshwater betaproteobacterium strain VA01 was isolated from Velka Amerika lake, Czech Republic, by the dilutionto-extinction technique (M. Mašín, unpublished data).…”

“…The distinctive fatty acids were also explored among different species (Koblížek et al, 2003;Yoon et al, 2004;Rontani et al, 2005). However, no comparison in phospholipid fatty acids (PLFA) has been made between BChl a-containing AAPB and its phylogenetically related bacteria lacking of BChl a.…”
