GATA3 induces mitochondrial biogenesis in primary human CD4+ T cells during DNA damage

Abstract: GATA3 is as a lineage-specific transcription factor that drives the differentiation of CD4+ T helper 2 (Th2) cells, but is also involved in a variety of processes such as immune regulation, proliferation and maintenance in other T cell and non-T cell lineages. Here we show a mechanism utilised by CD4+ T cells to increase mitochondrial mass in response to DNA damage through the actions of GATA3 and AMPK. Activated AMPK increases expression of PPARG coactivator 1 alpha (PPARGC1A or PGC1α protein) at the level of… Show more

“…We focused our attention on these proteins in the jejunum to examine their relationship with the mechanism regulating CD4+ T-cell differentiation, as well as the effects of LTA on these proteins. GATA3 promotes the differentiation of CD4+ T cells into Th2 cells . T-bet promotes the differentiation of CD4+ T cells into Th1 cells and releases IFN-γ, thereby regulating the inflammatory response .…”

“…S1, B to C). Of more importance to our study, genes that were decreased in expression in the presence of anti–TNF-α (and therefore whose expression is normally driven by TNF-α signaling) included several metabolic genes, including LDHA and SDHC , which are involved in glycolysis and the TCA cycle, respectively; GATA3 , which is essential for T cell differentiation and is also implicated in mitochondrial function ( 24 ); and MYC, which encodes the key transcription factor c-Myc that regulates the metabolic activity of T cells (Fig. 2B) ( 25 , 26 ).…”

“…Interestingly, mitochondrial reactive oxygen species could induce the activation of primary human T cells by regulating IL-2 and IL-4 expression [ 60 ]. GATA3, a cell-specific marker of TH2 cells, could modulate mitochondrial biogenesis and increase the mitochondrial content [ 61 , 62 ]. The abovementioned studies show that mitochondrial regulation, cell metabolism, and oxidative stress may play certain roles in the differentiation and activation of TH2 cells, which may be related to the difference in TH2 cell infiltration between the high- and low-risk groups.…”
