Gastroretentive raft liquid delivery system as a new approach to release extension for carrier-mediated drug

Abstract: Gabapentin (GBP), an antiepileptic and anti-neuropathic agent, suffers from short half-life (5–7 h), has narrow absorption window, and is absorbed via carrier-mediated mechanism resulting in frequent dosing, poor compliance, and poor bioavailability (<60%). Moreover, GBP is a freely water-soluble drug, thus it is considered a challenging candidate to be formulated as extended release dosage form. In this study, raft forming systems were investigated as a potential drug delivery system for prolonging gastric re… Show more

“…In vivo assessment was performed in rats to evaluate gastric residence of the gel formed. The increment in relative bioavailability of GBP from the optimized formula was 1.7 fold compared to commercially available Neurontin ® [158].…”

“…Thus, they can be used for the effective management of gastric esophageal reflux disease. When raft-forming systems come into contact with gastric fluid, they swell and form a viscous cohesive gel leading to the formation of a continuous layer termed as rafts [17,62]. Fabregas et al [89] explained the antacid raft-forming floating system.…”

“…Thus, CO 2 gas is generated that lowers the bulk density of the system, and as a result, the raft floats on the gastric fluid. Nabarawi et al [62] developed a controlled release floating raft system of mebeverine hydrochloride, and evaluated different excipients for their floating behavior and in vitro controlled-release. It forms a viscous and cohesive gel when it swells and entraps CO 2 bubbles produced by the reaction of carbonates and gastric fluid [12].…”

“…Various types of GRDDS with different gastroretentive mechanisms have been investigated, such as floating [4], high-density [5], muco-adhesive [6], raft-forming [7], magnetic [8], and expandable systems [9]. The most extensively explored types of GRDDS are floating or expandable systems.…”
