The present investigation evaluates the anti-stress activity of risperidone (RIS) in the cold restraint stress (CRS) model and related stress pathways. Rats were pretreated with RIS (0.1 and 1.0 mg/kg) for 21 days before subjecting to CRS. Ultra low dose of RIS (ULD; 0.1 mg/kg) in contrast to higher dose (1.0 mg/kg) significantly reduced stress in terms of ulcer index. ULD also reversed stress-induced increase in plasma corticosterone and norepinephrine levels used as markers for the function of hypothalamo-pituitary-adrenal (HPA) axis and sympathetic nervous system (SNS) respectively. ULD caused dose and brain region (hippocampus, prefrontal cortex and striatum) specific changes to stress-induced perturbations of serotonin, dopamine and its metabolites indicating modulation of brain monoaminergic system (BMS). ULD did not show any extrapyramidal side effects. Thus, the anti-stress effect ULD is probably mediated through the HPA axis, SNS and BMS. The study indicates a potential use of ULD in stress disorders.

В детском возрасте кислотные ожоги пищевода чаще всего встречаются в возрасте от 1 года до 8 лет. В месте локализации химической травмы увеличиваются процессы синтеза и деградации белков. Рубцовые изменения пищевода, развивающиеся в результате ожога, остаются одной из наиболее сложных проблем. Несмотря на многочисленные исследования, недостаточно изучены вопросы заживления и процессов рубцевания на клеточном и молекулярном уровне. Показаны количественные изменения белкового состава в условиях розвития кислотного ожога пищевода. Дальнейшее исследование процессов заживления может быть полезным в создании эффективных подходов для профилактики образования рубцовых изменений пищевода.Ключевые слова: ожог пищевода, белковые фракции, электрофорез. We investigated the involvement of central and peripheral D2 dopaminergic receptors in the mechanism of vascular permeability in rat's colon during experimental ulcerative colitis. Ulcerative colitis was induced in male white inbred rats by 6 % iodoacetamide enema. For the investigation of central and peripheral D2R, separate and joint injections of D2R antagonist domperidone (2 mg/100 g, per os) and D2R agonist quinpirole (1 mg/100 g, per os) were applied. Central D2R were destroyed by neurotoxin injection -6-OHDA. Colonic vascular permeability was measured by colonic extravasation of 1,5 % Evans blue. It was observed that blockade of peripheral D2R decreased colonic vascular permeability, while simultaneous activation of central D2R and inhibition of peripheral D2R have additive positive effect in prevention of increased colonic vascular permeability during experimental colitis.

Antipsychotic use is inversely associated with gastric cancer risk: A nationwide population‐based nested case‐control study

Objective The association between antipsychotic use and gastric cancer risk remains unclear. Therefore, this study aimed to determine the association between antipsychotic exposure and the incidence of gastric cancer. Methods Using a nested case‐control design, a total of 34 470 gastric cancer patients and 163 430 nongastric cancer controls were identified from Taiwan's National Health Insurance Research Database between 1 January 1997 and 31 December 2013. We analyzed the data using a conditional logistic regression model to adjust for possible confounding variables. Results Antipsychotic use was independently inversely associated with gastric cancer risk after controlling for potential confounding factors including income, urbanization, medications, physical and medical illness, aspirin use, nonsteroidal anti‐inflammatory drug use and triple therapy. In addition, dose‐dependent trends against gastric cancer risk were also shown with individual antipsychotic compounds including thioridazine, haloperidol, sulpiride, clozapine, olanzapine, quetiapine, amisulpride, and risperidone. A sensitivity analysis showed that second‐generation antipsychotics had significant dose‐dependent effects in reducing the risk of gastric cancer risk in patients with and without peptic ulcer disease. Conclusions Antipsychotic use was inversely associated with gastric cancer risk, and dose‐dependent effects against gastric cancer were also seen with several individual antipsychotic compounds.
