Gastric cancer is related to high mortality rates and advanced disease stage at the time of diagnosis. Its carcinogenesis is extensively studied and is associated with genetic and epigenetic changes, changed the interaction between tumor and adjacent stromal cells, and changes in the microenvironment molecule status. NEDD9 affects different signaling proteins and pathways, apoptosis, adhesion, cell migration, and invasiveness. Connexin-43 also assists in intercellular communications and has several channel-independent functions. Aberrant expression of those two gap junction proteins plays an essential role in metastatic processes. Our scope was to detect the expression of connexin-43 and NEDD9 in epithelial and stromal gastric cancer compartments and its relation to tumor progression and lymph node metastases. Cancer tissue from 53 cases of node-negative and 55 cases of node-positive primary gastric carcinoma patients was analyzed for connexin-43 and NEDD9 expression by immunohistochemical assay, and the results were correlated with the remaining clinical and pathological findings and survival. In our cohort of patients with lymph node metastases, we detected higher expression of epithelial connexin-43 in the primary tumor and stromal connexin-43 expression correlated with both epithelial NEDD9 (rho=0.453) and stromal NEDD9 (rho=0.484). Higher epithelial connexin-43 and NEDD9 expression were associated with higher mortality (HR 1.54, 95% CI 1.01 -2.37, p=0.048). Epithelial connexin-43 expression, both epithelial and stromal NEDD9 expression, T and N status were all independently associated with shorter survival. In summary, our findings suggest that increased expression of both epithelial and stromal NEDD9 and epithelial connexin-43 could potentially be used as prognostic gastric cancer biomarkers.

For explore the potential connection of RNA binding proteins (RBPs) to the expression function of gastric cancer (GC). We download the GPL10558 and GPL6947 platform mircroarray data from Gene Expression Omnibus (GEO) and Express database. Then the system integrates and analyzes the differentially expressed RBPs. And enrich the differentially expressed RBPs to understand the mechanism of its influence on tumors. Univariate Cox, lasso regression and multivariate Cox regression analysis were used to screen independent prognostic parameters to construct prognostic model, and calculate aera under time-dependent receiver operating characteristics (AUC) and survival analysis were used to evaluate their prognostic ability. GSE15459, GSE62254 cohorts were used to verify hub signature. Finally, we also verified the prognosis and expression of hub-RBPs. Systematic analysis identified 23 up-regulated and 30 down-regulated RBPs, and enrichment analysis showed that they mainly affect their modification by binding to mRNA, and their stability affects the progression of GC. After multiple statistical analyses, we obtained the prognostic signature constructed by 10 RBPs and determined that it has better predictive performance (AUC = 0.685). Through comprehensive bioinformatics analysis, we have obtained 10 key gastric cancer RBPs as potential prognostic biomarkers, providing new perspectives for the treatment and prognostic of GC.

Background: Gastric cancer (GC) is the most common malignant tumors in the world and surgical resection remains the primary treatment for it. Postoperative patients often suffer from gastrointestinal dysfunction as the most common side effects of surgery for GC patients. Acupuncture has a regulatory effect on gastrointestinal function. We conducted this study to assess the effectiveness of acupuncture on the restoration of gastrointestinal function of postoperative patients with GC. Methods: Seven electronic databases will be searched from inception to November 2020 to identify any relevant study: Medline, Embase, Cochrane Central Register of Controlled Trials (CENTRAL), China National Knowledge Infrastructure (CNKI), Wanfang Database, Chinese Biomedical Literature Database (CBM), and Chinese Scientific Journal Database (VIP database). No restriction on time and language. The primary outcome measure will be the Time to First Flatus and secondary outcome measures include the time of first defecation and the quality of life (QOL) and the number of patients with abdominal distention. We will use EndNote V.9.1 to screen the eligible literature and the I 2 statistic to assess heterogeneity in the included studies. The meta-analysis will be conducted using the Review Manager (RevMan) software (V.5.3). Results: Our study aims to systematically assess whether the pooled effects of currently available trials prove effects of acupuncture in improving gastrointestinal function of patients with GC in the postoperative period. Conclusion: This study will conduct an evaluation about the efficacy of acupuncture for the recovery of gastrointestinal function of patients with GC in the postoperative period, making up for the lack of relevant clinical evidence.
