Bacteriol. 85: 1365–1370. 1963.—Washed suspensions of Bacillus megaterium oxidized to CO 2 about 39% of the U-C 14 -glucose supplied and incorporated about 37% of the label by the time a marked break in the rate of O 2 consumption was noted. Almost one-half of the label was lost from the cells on acidification of the suspension. The remainder of the C 14 was present in the supernatant fluid, primarily in forms as yet unidentified, but other than carbohydrate. Both the Embden-Meyerhof and hexose monophosphate pathways of oxidation were involved. Endogenous respiration appeared to be inhibited only to a slight extent in the presence of an exogenous substrate. C 14 appeared in all fractions of the cells; the highest percentage of firmly bound C 14 was present in hot 5% trichloroacetic acid-insoluble matter. A decrease in C 14 content of the various fractions was noted during endogenous respiration of cells labeled during growth. Pyruvate and acetate were oxidized very slowly by B. megaterium . The results indicate the complexity of oxidative assimilation and the dynamic state of cellular metabolism.

1. Extracts prepared from a variety of higher-plant tissues by ammonium sulphate fractionation were shown to catalyse the interconversion of glycine and serine. This interconversion had an absolute requirement for tetrahydrofolate and appeared to favour serine formation. 2. The biosynthesis of serine from glycine was studied in more detail with protein fractionated from 15-day-old wheat leaves. Synthesis of [(14)C]serine from [(14)C]glycine was not accompanied by labelling of glyoxylate, glycollate or formate. 3. The synthesis of serine from glycine was stimulated by additions of formaldehyde, and [(14)C]formaldehyde was readily incorporated into C-3 of serine in the presence of tetrahydrofolate. 4. The results are interpreted as indicating that serine biosynthesis involves a direct cleavage of glycine whereby the alpha-carbon is transferred via N(5)N(10)-methylenetetrahydrofolate to become the beta-carbon of serine.

The composition of the insoluble "integuments" and soluble "contents" fractions of spores of four Bacillus species of widely differing heat resistance were compared. Electron microscopy of thin sections was also used to determine and compare the morphological structures in the integument preparations. The soluble fractions of the thermophiles, B. coagulans and B. stearothermophilus, had a higher content of hexose and dipicolinic acid. The hexose content of both fractions of the four species was related to heat resistance. Integument fractions consisted chiefly of protein together with variable amounts of the mucopeptide constituents, a, e-diaminopimelic acid (DAP) and hexosamine. In the thermophiles the DAP and hexosamine were found chiefly in the insoluble integuments fractions, while in B. cereus and B. subtilis most of this material was soluble. Integument preparations, containing mainly protein with little mucopeptide, consisted chiefly of outer and inner spore coats, while preparations having more mucopeptide contained also residual cortical material and a cortical membrane (possibly the germ cell wall). The results suggest that spore integuments consist of mainly proteinaceous outer and inner coats together with variable amounts of residual cortex and cortical membrane which contain the mucopeptide material.
