Gap junctions: structure and function (Review)

Abstract: Gap junctions are plasma membrane spatial microdomains constructed of assemblies of channel proteins called connexins in vertebrates and innexins in invertebrates. The channels provide direct intercellular communication pathways allowing rapid exchange of ions and metabolites up to approximately 1 kD in size. Approximately 20 connexins are identified in the human or mouse genome, and orthologues are increasingly characterized in other vertebrates. Most cell types express multiple connexin isoforms, making like… Show more

“…The density of functional Cxs on the surface membrane can be influenced by modulation of transport along cytoskeletal elements, and a gap junction's resident time in the surface membrane is likely to be short because of the rapid turnover of Cxs (1-5 h) (18,30). Thus Cx mimetic peptides are usually exposed to the tissues at least for 1 h to block not only the Cxs in the surface membrane but also renewed Cxs from cytosol.…”

“…Ions or small molecules pass through these critical intercellular pathways to regulate various physiological and developmental processes. 5 The GJ channels are double-membrane protein structures that form by head-to-head docking of two half channels, termed connexons, to create hydrophilic pores across the membranes. 6 Each connexon is composed of six polytopic transmembrane protein subunits, termed connexins.…”

“…Establishment of gap junctions affects a number of physiological functions such as cell growth, proliferation, and differentiation (Evans & Martin 2002). We showed that formation of cell-to-cell contacts between thyroid cancer cells in low-adherent condition was associated with decreased proliferation.…”
