Gangliosides inhibit the development from monocytes to dendritic cells

Abstract: SUMMARYDendritic cell (DC) development and function is critical in the initiation phase of any antigen-specific immune response against tumours. Impaired function of DC is one explanation as to how tumours escape immunosurveillance. In the presence of various soluble tumour-related factors DC precursors lose their ability to differentiate into mature DC and to activate T cells. Gangliosides are glycosphingolipids shed by tumours of neuroectodermal origin such as melanoma and neuroblastoma. In this investigatio… Show more

“…At the time of hsp loading, immature DCs expressed CD91 (data not shown) known to be a receptor for efficient uptake of gp96 (16). We have documented the endocytotic activity of this DC population by showing the successful uptake of FITC-dextran (55). In addition, our previous observation that the type of DCs used in our system can present recombinant antigenic protein to HLA class II-restricted Ag-specific T cell clones indicates that these DCs have an intact Agprocessing capacity (37).…”

“…Incubation of human monocytes with GM2 inhibits the differentiation of DC in the presence of cytokines, as demonstrated by enhanced adherence and cell spreading with reduced expression of MHC class II and costimulatory molecules; in contrast, GM3 had no effect (51). In a more recent study looking at the effects of human milk-derived gangliosides, GD3 inhibited maturation of bone marrow-derived DC in response to LPS, as measured by diminished production of proinflammatory cytokines and reduced proliferation in a MLR, whereas GM3 had lesser effects (52).…”

“…Wölfl et al (38) using commercially available bovine brain ganglioside mixture. In that study, however, the inhibition could only be related to GM2, whereas GM3 and GD3 had no effect.…”
