Gamma oryzanol impairs alcohol-induced anxiety-like behavior in mice via upregulation of central monoamines associated with Bdnf and Il-1β signaling

Abstract: Adolescent alcohol exposure may increase anxiety-like behaviors by altering central monoaminergic functions and other important neuronal pathways. the present study was designed to investigate the anxiolytic effect of 0.5% γ-oryzanol (GORZ) and its neurochemical and molecular mechanisms under chronic 10% ethanol consumption. Five-week-old ICR male mice received either control (14% casein, AIN 93 M) or GORZ (14% casein, AIN 93 M + 0.5% GORZ) diets in this study. We showed that GORZ could potentially attenuate a… Show more

“…Araujo et al reported that it had antidepressant-like effects in Drosophila melanogaster exposed to chronic unpredictable mild stress 17 . Additionally, Akter et al demonstrated that ORY exhibits anxiolytic effects by ameliorating acute stress-induced behavioral anxiety in mice and attenuates alcohol-induced anxiety via the upregulation of monoamines, BDNF, and IL-1β signaling 38,39 . These ndings are in line with our results, indicating that ORY treatment effectively alleviated depressive-like behaviors in mice exposed to CRS.…”

“…During binge drinking, it is possible that protective effects on cognition and neuroinflammation could be achieved by potentiating the endocannabinoid anandamide (AEA) neurotransmission [ 263 ]. As AEA is metabolized by the specific enzyme fatty acid amide hydrolase (FAAH), selective inhibitors of this enzyme might have therapeutic potential [ 264 , 265 ].…”

“…Studies have shown that only 100 mg/kg/day, γ-oryzanol can modulate the hippocampal protein that is involved in synaptic and neuronal signaling, thereby functioning as a neuroprotector and antioxidant. On the other hand, 0.5% γ-Oryzanol can control anxiety-like behavior by modulating the monoaminergic neuronal signaling pathway 73,74) . Thus, the bioactive component of RBO, γ-oryzanol, exhibits its candidature as a therapeutic moiety to improve cognitive behavior in neurodegenerative disorders.…”
