Gallium-68-labeled DOTA-rhenium-cyclized α-melanocyte-stimulating hormone analog for imaging of malignant melanoma

Abstract: Introduction-Diagnosis of malignant melanoma is critical, since a patient's prognosis is poor. Previous studies have shown that 64 Cu-and 86 Y-DOTA-ReCCMSH(Arg 11 ) have the potential for early detection of malignant melanoma by exploiting the sensitivity and high resolution of PET. This encouraged us to investigate DOTA-ReCCMSH(Arg 11 ) labeled with another β + -emitting radionuclide, 68 Ga.Methods-DOTA-ReCCMSH(Arg 11 ) was successfully labeled with 68 Ga at pH 3.8-4 at 85 ºC. Acute biodistribution and small … Show more

“…15,27,28 Hence, 15 mg of L -lysine was co-injected with either 99m Tc-RGD-Lys-(Arg 11 )CCMSH or 99m Tc-RGD-Arg-(Arg 11 )CCMSH at 2 h post-injection to determine whether the non-specific renal uptakes of 99m Tc-RGD-Lys-(Arg 11 )CCMSH and 99m Tc-RGD-Arg-(Arg 11 )CCMSH were associated with the electrostatic interaction between the peptide and kidney cells. As we anticipated, co-injection of L -lysine reduced the renal uptake of 99m Tc-RGD-Lys-(Arg 11 )CCMSH by 52.1% (p<0.05) and decreased the renal uptake of 99m Tc-RGD-Arg-(Arg 11 )CCMSH by 27.7% (p<0.05) without affecting the tumor uptakes, demonstrating that the electrostatic interaction played an important role in the non-specific renal uptakes of 99m Tc-RGD-Lys-(Arg 11 )CCMSH and 99m Tc-RGD-Arg-(Arg 11 )CCMSH.…”

“…Up to now, many variants based on α-MSH and its biologically stable synthetic analog Ac-[Nle 4 , DPhe 7 ]-α-MSH (NDP-MSH), which binds to all melanocortin receptors except for MC2R with similar affinity, has been synthesized [100]. Among them are linear and cyclic (for fixation the most favorable conformation for receptor binding) peptides conjugated with chelators for 111 In [101110], 99m Tc [101, 109, 111115], and 67 Ga [116] for diagnostics using single photon emission computed tomography (SPECT), and also for 18 F [117119], 68 Ga [110, 116, 120, 121], 64 Cu [122, 123], and 86 Y [122] for positron emission tomography (PET) diagnostics. Gene therapy using Na-I symporter (NIS) is another approach exploiting MC1R–specific ligands [124].…”

“…A group of investigators at Washington University in St. Louis, Missouri, reported tumor uptake in mice as well as the biodistribution of a 68 Ga-labeled DOTA-rhenium-cyclized a-MSH analog (24). This agent showed promise for the early detection of malignant melanoma.…”
