Galectins‐1 and ‐3 in Human Intervertebral Disc Degeneration: Non‐Uniform Distribution Profiles and Activation of Disease Markers Involving NF‐κB by Galectin‐1

Abstract: Degeneration of the human intervertebral disc (IVD) is assumed to underlie severe clinical symptoms, in particular chronic back pain. Since adhesion/growth‐regulatory galectins are linked to arthritis/osteoarthritis pathogenesis by activating a pro‐degradative/‐inflammatory gene expression signature, we hypothesized a similar functional involvement of galectins in IVD degeneration. Immunohistochemical evidence for the presence of galectins‐1 and ‐3 in IVD is provided comparatively for specimens of spondylochon… Show more

“…Interestingly, we found the expression of galectin-3 was significantly upregulated in the H 2 O 2 -induced NP cells. As previous reported, galectin-3 was aberrantly expressed in IDD [ 7 , 10 , 36 ] and inhibition of galectin-3 alleviates the spinal cord injury and IDD [ 7 , 8 ]. Galectin-3 is involved in two coordinated pathways of programmed cell death, apoptosis, and autophagy [ 37 ], suggesting that galectin-3 may be involved in the H 2 O 2 -induced apoptosis and autophagy of NP cells.…”

“…Tissue and organoid specimens were fixed in 7% paraformaldehyde, dehydrated in ethanol and paraffinembedded according to standard in-house procedures. 35 Paraffin sections of approx. 2.5 μm thickness were stained with hematoxylin and eosin (HE) for morphological evaluations.…”

“…RNA was reverse transcribed into cDNA (Firma und Kit einfügen) and subsequent RT-qPCR was performed on a QuantStudio3 QPCR system (Applied Biosystems, Thermo Fisher) using SensiMix SYBR Green Master Mix as previously described. 35 Primer sequences are described in ESI † Table S3.…”

“…In-Cell Western (ICW) assay. The ICW assay was performed using the Odyssey Imaging System (LI-COR Biosciences) according to a previously described protocol (Elshamly et al 2019 ). In brief, chondrocytes were grown in 96-well plates, and starved cells underwent Gal-4 (50 µg/ml) treatment for 1 h. Cells were washed and fixed with methanol (–20 °C) for 10 min.…”
