Galectin signature of the choriocarcinoma JAr cells: Galectin‐1 as a modulator of invasiveness in vitro

Abstract: Our previous findings showed that galectin-1 (LGALS1) plays an important role in the in vitro invasion of normal human trophoblast cells. In the present study, choriocarcinoma JAr cells were found to express LGALS1, -2, -3, -8, -10, and -13 mRNA and at least LGALS1, -3, and -8 protein, as determined by reverse-transcriptase PCR and Western blot, respectively. The galectin mRNA signature of JAr cells thus differed from that of normal first-trimester extravillous trophoblasts. A Matrigel migration assay was also… Show more

“…Among trophoblast cells, galectin-3 was localised in villous cytotrophoblast, and the cytotrophoblast of middle and distal cell columns of anchoring villi 10 . Expression of this galectin was also reported in freshly isolated trophoblast cells, extravillous HTR-8/SVneo cells, BeWo, JAr and JEG-3 choriocarcinoma cells 10,11,23 . It has been reported that galectin-3 can be associated with some pathological conditions involving the trophoblast, such as preeclampsia, HELLP syndrome and trophoblast malignancies 24,25 .…”

“…Expression of galectin-3 has been previously documented for villous cytotrophoblast, cell columns, isolated cytotrophoblast and trophoblast derived cell lines 10,11,23 . Here, the expression pattern and subcellular distribution of galectin-3 in HTR-8/SVneo cells was further examined using polyclonal anti-galectin-3 antibodies.…”

“…The research from our lab has previously shown that galectin-1 may act as a part trophoblast invasion machinery [41]. Both oxidized and reduced forms of recombina human galectin-1 (rhgal-1) stimulated first-trimester CTB and HTR-8/SVneo cell invasio This stimulation was reversed by neutralizing antibodies, as was by treatment with inhi itory sugar lactose, suggesting a lectin-type interaction-dependent mode of stimulatio These observations were replicated for the JAR trophoblast cell line, as well [42]. Prolife ation of BeWo cells was inhibited by the treatment with galectin-1 [43].…”

“…The data presented show that all three analyzed gals were released from HTR-8/SVneo cells, as assessed by western blot of different fractions of CM. Our group previously demonstrated that gal-1 is secreted by HTR-8/SVneo cells (Ćujić et al, 2013) and the presence of all three galectins in CM of the choriocarcinoma cell line JAr (Kolundzic et al, 2015). Secreted gal-3 was detected in CM from another choriocarcinoma cell line, BeWo (Yang et al, 2014).…”

“…Gal-1, gal-3, and gal-8 have been detected in conditioned medium (CM) of trophoblast cell lines (Yang et al, 2011;Ćujić et al, 2013;Kolundzic et al, 2015). As gals lack a signal sequence, questions pertinent to the route of gal secretion in trophoblasts remain unresolved (Hughes, 1999).…”
