Galectin-9 suppresses tumor metastasis by blocking adhesion to endothelium and extracellular matrices

Abstract: We previously described an inverse correlation between galectin-9 (Gal-9) expression and metastasis in patients with malignant melanoma and breast cancer. This study verified the ability of Gal-9 to inhibit lung metastasis in experimental mouse models using highly metastatic B16F10 melanoma and Colon26 colon cancer cells. B16F10 cells transfected with a secreted form of Gal-9 lost their metastatic potential. Intravenous Gal-9 administration reduced the number of metastases of both B16F10 and Colon26 cells in t… Show more

“…In addition, we have also seen suppression of VLA-4 (␣-4/␤-1)-VCAM-1 interactions and suppression of metastasis in a murine model of cancer. 44 Thus, the function of Gal-9 could be contextual and different depending on the microenvironment, and the physiologic concentration of Gal-9 in different microenvironments in vivo is not known.…”

“…Association of Gal-1 with ECM proteins causes a direct reduction in leukocyte adhesion, as well as inhibiting T-cell migration through the ECM (He & Baum 2006), whereas Gal-3 localisation with ECM enhances leukocyte adhesion. Gal-9 has also been shown to reduce tumour cell adhesion by preventing binding to ligands on the endothelium and ECM (Nobumoto et al 2008).…”

“…[4][5][6][7][8][9][10] Moreover, recent studies have indentified anti-cancer properties of galectin-9 against several cancers. [11][12][13][14][15] In this study, we show the anti-multiple myeloma (MM) activity of a recombinant mutant form of human galectin-9 (hGal9) through the activation of c-Jun NH 2 -terminal kinase (JNK) and p38 mitogen-activated protein kinase (MAPK) signaling pathways, those share crucial roles in the survival and death of myeloma cells. 16 …”
