Galectin-1 stimulates motility of human umbilical cord blood-derived mesenchymal stem cells by downregulation of smad2/3-dependent collagen 3/5 and upregulation of NF-κB-dependent fibronectin/laminin 5 expression

Abstract: Galectin-1 (Gal-1) belongs to a family of endogenous lectins with conserved carbohydrate recognition domains binding β-galactosidase sugars and plays a vital role in regulating stem cell functions including determination of cell fate. However, our understanding of the functional roles of Gal-1 in human umbilical cord blood-derived mesenchymal stem cells (UCB-MSCs) is still fragmentary and incomplete. Gal-1 significantly increased motility after a 24-h incubation, and this effect was inhibited by β-lactose. We … Show more

“…Although the discrepancy with regard to calcium influx may due to differences in species, cell types, or experimental conditions, our results indicate that motility of UCB-MSCs, completely abrogated by the inhibition of PKC pathways, provide strong evidence that PKC plays an important role in the Ntn-1 signaling pathway. These results are consistent with previous study in which galectin-1 increased PKC phosphorylation, but did not affect calcium influx in promoting UCB-MSCs motility [30]. We further found that PKCa, one of several major PKC isoforms, is uniquely activated by Ntn-1.…”

“…Laminin-332 is a migration factor for human corneal epithelial cells, where it is active as both a chemoattractant and haptotactic factor. [9][10][11][63][64][65][66] As mentioned above, laminin-332 mRNA and protein are expressed at the leading edge of the migrating tongue in wounded epidermis. 12 It is not known exactly how Tb4 induces laminin-332, but it has been shown that Tb4 stabilizes HIF1, which is a transcription factor that binds to the promoter of the a3 chain of laminin-332.…”

“…M17 broth was supplemented by 10% lactose to provide sufficient carbohydrates for the growth of EcN. A study has reported that lactose inhibited the motility of human umbilical cord bloodderived mesenchymal stem cells (41). High lactose containing media may have been responsible for the toxic effect of M17 broth and EcN SN (M17C) to Caco-2 cells (41).…”
