GAL4 enhancer traps expressed in the embryo, larval brain, imaginal discs, and ovary of drosophila

Abstract: We have screened a collection of approximately 400 GAL4 enhancer trap lines for useful patterns of expression in the embryo, larval brain, imaginal discs, and ovary using a UAS‐lacZ reporter construct. Although similar patterns of expression have previously been reported in the original P[lacZ] enhancer trap screens, these lines are useful for directing ectopic expression of genes in discrete patterns during these stages. In addition, we have identified some unique patterns of expression that have not been pre… Show more

“…The dorsal closure defects of tkv mutant embryos can be significantly suppressed by amnioserosa-specific expression of Tkv Q199D and we wondered if similarly overexpressing DACK in this tissue alone would suppress Dpp pathway defects, particularly considering that global activation of Cdc42 signaling causes an up-regulation of DACK expression specifically in the amnioserosa (Sem et al, 2002;Fernandez et al, 2007). Expression of DACK using the amnioserosa-specific driver Gal4 c381 (Manseau et al, 1997) suppressed the defects of tkv 7 embryos by roughly 30% (Table 1). To see if DACK function had any bearing on zip transcription in the amnioserosa, we expressed wild-type and kinasedead (KD) DACK transgenes (Sem et al, 2002) using Gal4 c381 and performed FISH.…”

“…Systematic screening for enhancers and suppressors of PEV identified many important genes that modulate chromatin compaction and gene silencing (9). Genes expressed using the bipartite GAL4/UAS system also show variable expression levels (10)(11)(12)(13). The basis of GAL4/UAS variegation has remained obscure, but we wondered if it could be used to assay epigenetic stability.…”

“…For the RNAi-mediated interference, we used the tsl(G)GAL4, 198Y, c306, c522, and cb07 lines expressed in the BCs and the tsl(F)GAL4, cb35, 55B, cb41, and dppGAL4 expressed in the CCs. For a description of these lines, see the following sources: tsl(G)GAL4 and tsl(F)GAL4 (see below), 198Y, c306 and c522 (29), cb07, cb35 and cb41 (30), and 55B (28,31). We have examined egg chambers from flies carrying the 55B line, a UAS-GFP construct and the tsl(G)-Bgal construct, which is specific for the BCs and PFCs at the confocal and in no case is there an overlap between the two domains (n ϭ 14) (data not shown).…”
