GABA transport in the rat thyroid

Abstract: 1. The uptake of gamma-aminobutyric acid (GABA) into rat thyroid slices was studied. 2. Uptake of 14C-GABA was concentration-dependent: one unsaturable (diffusion) and two saturable components obeying Michaelis-Menten kinetics contributed to transport. 3. The kinetic constants of saturable GABA transport systems were: Km1 = 1.5 microM, V1 = 4.0 nmol x (g wet weight)-1 x (20 min)-1 (high-affinity uptake): Km2 = 800 microM, V2 = 260 nmol x (g wet weight)-1 x (20 min)-1 (low-affinity uptake). 4. Uptake mediated b… Show more

“…Kinetic analysis indicates that GABA is transported by a saturable uptake process. The mechanism of 3H-GABA uptake in the ante rior pituitary was of lower efficiency than in cerebral cor tex [14], However, kinetic parameters of 3H-GABA up take in the anterior pituitary are in agreement with data reported for nonneuronal tissues [12]. Anterior pituitary 3H-GABA uptake was sensitive to the inhibition pro duced by the GABA-related compounds that are known to inhibit neuronal and/or glial GABA uptake [19][20][21].…”

“…These results show that the anterior pituitary gland can accumulate 3H-GABA in a temperature-and sodiumdependent manner, and this was similar to the GABA uptake by other tissues [12,14,17,18]. Kinetic analysis indicates that GABA is transported by a saturable uptake process.…”

“…GABA appears to play a role in the homeostasis of the normal thyroid [13]. The thyroid gland has been shown to both actively take up and synthesize GABA [14,15]. Research in rodent models suggests a complex interaction between thyroid hormone and GABA, with each regulating secretion or uptake of the other.…”
