hWe explored the use of matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) for identification of Fusobacterium nucleatum subspecies. MALDI-TOF MS spectra of five F. nucleatum subspecies (animalis, fusiforme, nucleatum, polymorphum, and vincentii) were analyzed and divided into four distinct clusters, including subsp. animalis, nucleatum, polymorphum, and fusiforme/vincentii. MALDI-TOF MS with the modified SARAMIS database further correctly identified 28 of 34 F. nucleatum clinical isolates to the subspecies level. Fusobacterium nucleatum is an opportunistic pathogen, associated with various forms of periodontal diseases and extraoral infections, as well as colorectal cancer (1, 2). F. nucleatum is a highly heterogeneous species and was classified into five subspecies: animalis, nucleatum, polymorphum, vincentii, and fusiforme (3-5). Recent studies based on phylogenetic analysis of the nucleic acid sequences of 16S rRNA, rpoB, zinc protease, and 22 other housekeeping genes suggested that F. nucleatum subsp. fusiforme and vincentii be classified into a single subspecies, F. nucleatum subsp. fusiforme/vincentii (4, 5).Different subspecies may vary in pathogenesis relating to different levels of disease activity (6-8). Fusobacterium nucleatum subsp. nucleatum is isolated mostly in periodontal diseased sites, whereas F. nucleatum subsp. fusiforme/vincentii is often isolated from healthy sites as normal flora (9). F. nucleatum subsp. animalis and polymorphum are associated with pregnancy complications (7), and F. nucleatum subsp. animalis is also associated with inflammatory bowel disease (10). Until now, molecular technologies have been the most effective and widely accepted tools for subspecies identification (3,4,11). At the subspecies level of F. nucleatum, the sequence divergences of 16S rRNA genes were only 0.6% to 1.9%, so full-length sequencing of 16S rRNA was desirable (4).This study aimed to explore the use of matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) for the identification of F. nucleatum at the subspecies level. A commercially available database was amended using 15 F. nucleatum isolates comprising the type strains and other wellcharacterized clinical isolates of the five subspecies and then tested for rapid identification of F. nucleatum subspecies.(This study was presented in part at the 113th (10). Accuracy of commercial kits and published primer pairs for the detection of periodontopathogens

ObjectivesDespite the input of microbiome research, a group of 20 bacteria continues to be the focus of periodontal diagnostics and therapy. The aim of this study was to compare three commercial kits and laboratory-developed primer pairs for effectiveness in detecting such periodontopathogens.Materials and methodsFourteen bacterial mock communities, consisting of 16 randomly assembled bacterial strains, were used as reference standard for testing kits and primers. Extracted DNA from mock communities was analyzed by PCR in-house with specific primers and forwarded for analysis to the manufacturer’s laboratory of each of the following kits: ParoCheck®Kit 20, micro-IDent®plus11, and Carpegen® Perio Diagnostik.ResultsThe kits accurately detected Fusobacterium nucleatum, Prevotella intermedia/Prevotella nigrescens, Parvimonas micra, Aggregatibacter actinomycetemcomitans, Campylobacter rectus/showae, Streptococcus mitis, Streptococcus mutans, and Veillonella parvula. The in-house primers for F.nucleatum were highly specific to subtypes of the respective periopathogen. Other primers repeatedly detected oral pathogens not present in the mock communities, indicating reduced specificity.ConclusionsThe commercial kits used in this study are reliable tools to support periodontal diagnostics. Whereas the detection profile of the kits is fixed at a general specificity level, the design of primers can be adjusted to differentiate between highly specific strains. In-house primers are more error-prone. Bacterial mock communities can be established as a reference standard for any similar testing.Clinical relevanceThe tested kits render good results with selected bacterial species. Primers appear to be less useful for routine clinical diagnostics and of limited applicability in research. Basic information about the periodontopathogens identified in this study supports clinical decision-making.

Fusobacterium hwasookii sp. nov., Isolated from a Human Periodontitis Lesion

In this study, we classified the five strains (ChDC F128(T), ChDC F145, ChDC F174, ChDC F206, and ChDC F300) as a novel species of genus Fusobacterium by DNA-DNA hybridization and multi-locus phylogenetic analysis (MLPA), based on a single sequence (24,715 bp) of 22 concatenated housekeeping genes, with morphological and chemotaxonomic characteristics. DNA-DNA hybridization data showed that the values of genomic relatedness between ChDC F128(T) and each of the other novel strains were ranged from 79.0 to 82.6 %, while those of genomic relatedness between ChDC F128(T) and type strain of each of subspecies of F. nucleatum or Fusobacterium periodonticum were ranged from 40.9 to 54.4 %. MLPA revealed that the 5 strains were clustered as one group and clearly discriminated with F. nucleatum and F. periodonticum with 100 % bootstrap value. The DNA G+C content of the five novel strains were ranged from 26.9 to 27.0 mol%. The cellular fatty acid analysis of clinical isolates and type strains revealed C14:0, C16:0, and cis-9 C16:1 as the major fatty acids. The cell wall peptidoglycan of the 5 strains was comprised of meso-lanthionine. These results show that the 5 strains are novel species and belong to the genus Fusobacterium. Strain ChDC F128(T) (=KCOM 1249(T) = KCTC 5108(T) = JCM 30218(T)) is suggested to be the type strain of a novel species of genus Fusobacterium, for which the name Fusobacterium hwasookii sp. nov. is proposed.
