It was reported previously that proteolytic enzyme-treated M red cells lost their reactivity completely with only anti-M IgG antibody.In this paper, M red cells treated with trypsin for 45 min or 3 hr were used to prepare group-specific anti-M IgG and -N IgG antibodies as an adsorbent.It was revealed that M red cells treated for 3 hr could adsorb anti-N antibody in anti-M IgG fraction obtained from crude anti-M serum by DEAEcellulose column chromatography.The potency of the red cells to adsorb anti-N IgG antibody was similar to that of normal N red cells. Anti-N IgG antibody was prepared by heat-eluting trypsin-treated M red cells sensitized with anti-N fraction.However, the experiments with M red cells treated for 45 min showed unsatisfactory results.It was due to the red cells which could still react with high-titered anti-M IgG antibody but not sufficiently with anti-N IgG antibody.-MN blood groups; IgG; proteolytic enzymeThe MN blood grouping has been usually performed with rabbit anti-M and -N sera in which non-specific agglutinins were adsorbed by the red cells lacking the respective agglutinogens. Although anti-M and -N agglutinins from human, animals other than rabbit and plant seed are practically used, it has been pointed out that these agglutinins still contain some non-specific agglutinins (Race and Sanger 1968). In the previous paper (Sagisaka et al. 1972a), it was revealed that proteolytic enzyme-treated M red cells lost their reactivity completely with anti-M IgG antibody but became agglutinable with each of anti-M IgA and IgM antibodies and anti-N IgG, IgA and IgM antibodies. These findings suggested that trypsintreated M red cells could adsorb non-specific agglutinins in anti-M IgG and anti-N IgG fractions prepared from crude anti-M and -N sera by DEAE-cellulose column chromatography.In this paper, attempts to specify anti-M IgG fraction by adsorption and anti-N IgG fraction by adsorption-elution method using M red cells treated sufficiently with trypsin were described.
