BackgroundIdentifying the molecular interactions using bioinformatics tools before venturing into wet lab studies saves the energy and time considerably. The present study summarizes, molecular interactions and binding energy calculations made for major structural protein, collagen of Type I and Type III with the chosen cross-linkers, namely, coenzyme Q10, dopaquinone, embelin, embelin complex-1 & 2, idebenone, 5-O-methyl embelin, potassium embelate and vilangin.ResultsMolecular descriptive analyses suggest, dopaquinone, embelin, idebenone, 5-O-methyl embelin, and potassium embelate display nil violations. And results of docking analyses revealed, best affinity for Type I (- 4.74 kcal/mol) and type III (-4.94 kcal/mol) collagen was with dopaquinone.ConclusionsAmong the selected cross-linkers, dopaquinone, embelin, potassium embelate and 5-O-methyl embelin were the suitable cross-linkers for both Type I and Type III collagen and stabilizes the collagen at the expected level.

With the Teahnical Assistance of Richard A. MurrayThe effects of solutions of homogentisic acid and its analogues on hide powder collagen preparations have been studied by a volumetric technique. The data indicate that unoxidized homogentisic acid (HGA) reacts reversibly with collagen by pH-dependent electrovalent forces. Autoxidized, polymerized homogentisic acid solutions (HGA-OP), on the other hand, form irreversible linkages to collagen which are quite independent of pH. These are primarily of the coordinate, nonionic type and result in crosslinking of adjacent collagen polypeptide chains with increased stability of the collagen lattice (tannage).Solutiones de acido homogentisic monoe polymere esseva passate a transverso columnas chromatographic de isoelectric e n-acetylate collageno de pelle in pulvere. Esseva obtenite nulle indication de un ligation del non-oxydate monomer0 a1 collageno. Tamen, solutiones de autoxydate acido polymero-homogentisic (HGA-OP) esseva adsorbite fortemente. Le datos supporta le interpretation que HGA-OP es un potente agente tannante pro collageno de pelle in pulvere.WELLING IN aqueous solutions or in solutions of acids and bases is a S characteristic property of the sclero-protein collagen. The phenomenon involves a partial permanent disorganization of the collagen crystal lattice and is primarily due to a disruption of the stabilizing lateral links of the collagen For the most part, these stabilizing links are either longrange ionic, electrovabncy forces (-100 A ) or short-range coordinate forces of the hydrogen bridge and dipole attraction types (-5 A) or of the van der Waals types ( -3 A ) .4Where swelling arises principally in consequence of an electrovalent interaction between the swelling agent and the strongly ionic charged groups of the protein, osmotic or Donnan-Gibbs swelling results. In the presence of the primarily mokm2ar interactions (electrostatic) between the swelling agent and the nonionic groups of collagen, the resulting volume change of the solid phase ( lyotropic swelling) is, in contradistinction to Donnan-Gibbs swelling, more or less permanent even after removal of the dispersion medium.Compounds like acetic acid and certain phenols, which possess strong coordinate reactivity and which exist in solution either as hydrogen-bonded dimers6 or covalently-active dipoles,6 swell collagen principally lyotropically ( although in part osmoticalIy as well) by interacting with the ketoimide groups of adjacent collagen peptide linkagesna It would not be unreasonable, therefore,
